一株侵染白星花金龟幼虫的绿僵菌分离鉴定及培养条件初步研究  

Isolation and Identification of a Strain of Metarhizium sp.from Potosia brevitarsis Lewi Larvae and Preliminary Study of Its Culture Condition

摘  要:为弄清从昆明市云南农业大学后山马铃薯田自然罹病的白星花金龟( Potosia brevitarsis Lewi)幼虫虫体上获得的1株绿僵菌 ( Metarhizium sp.)的分类地位, 采用形态学特征和ITS-rDNA序列分析相结合的方法进行种类鉴定。 通过测量该菌株在SDAY、 PDA 两种培养基上的菌落直径和产孢量, 初步明确了该菌株的培养条件。鉴定结果表明该病原真菌为罗伯茨绿僵菌( Metarhizium robertsii ), 编号为Ma20191123菌株。此菌株在(25±1) ℃、 相对湿度80%、 光照16L∶8D的条件下利用SDAY、 PDA两种培养基接种后均能正常生长, 菌落直径分别为(2.56±0.15)、 (2.36±0.17) cm, 单位面积产孢量分别为(1.66± 0.25 )×10^(7)、 (1.24±0.47)×10^(7) 孢子/cm 2。该研究为蛴螬生物防治提供了候选菌种资源。To determine the taxonomic status for a strain of Metarhizium sp. isolated from infected cadaver of Potosia brevitarsis Lewi larvae from potato fields nearby Yunnan Agricultural University in Kunming City, Yunnan Province, the species identification was conducted by using morphological characterization and ITS-rDNA sequence analysis, and culture condition was preliminarily studied through determination of colony diameter and sporulation on different media. The results indicated the fungus was identified to be Metarhizium robertsii , which was named as Ma20191123. This strain could be grown on both SDAY and PDA media under culture condition of (25±1) ℃, relative humidity of 80% and photoperiod of 16 h light and 8 h darkness. The colony diameter and spore yield per area was (2.56±0.15) and (2.36±0.17) cm, (1.66± 0.25) × 10^(7) and (1.24±0.47) × 10^(7) spores/cm 2, respectively, on these two medias. The results of this study provided promising candidate strain for use in microbial control of white grubs.
