摘 要:HSP70是热激蛋白家族重要成员之一,在植物中广泛存在,并在植物非生物胁迫响应中发挥重要作用。为研究烟草NtHSP70-2基因的结构与功能,本研究对NtHSP70-2基因的启动子序列、蛋白理化性质和结构以及对温度胁迫的响应情况进行了分析。结果表明:NtHSP70-2基因编码完整的开放阅读框为1947 bp,编码649个氨基酸;编码蛋白的理论等电点为5.13,不稳定系数为35.03,分子量为71.10 kDa,预测为亲水性蛋白;NtHSP70-2与辣椒、番茄、马铃薯、茄子的相应同源序列相似度高;二级结构主要为α-螺旋,三级结构与辣椒CaHSP70的相似;NtHSP70-2主要含有P-box、CGTCA-motif、ABRE、STER、MYB、DRE core等顺式作用元件,这些元件都能响应温度胁迫。qRT-PCR分析结果表明,高低温胁迫12 h内,NtHSP70-2基因的相对表达量增加,胁迫12 h后,表达量逐渐减低;NtHSP70-2基因在烟草根、茎、叶中均有表达,叶中表达量高于茎和根中,相同部位高温胁迫下的相对表达量高于低温胁迫下的。综上,NtHSP70-2基因能响应温度胁迫,提高烟草对温度的耐受性。HSP70 is an important member of the heat shock protein family,which is widely present in plants and plays an important role in plant abiotic stress response.In order to study the structure and function of NtHSP70-2 gene in tobacco,the promoter sequence,protein physicochemical properties and structure were analyzed in this study,and its response to temperature stress was analyzed.The results showed that the NtH-SP70-2 gene encoded a complete open reading frame of 1947 bp,and encoded 649 amino acids.The NtH-SP70-2 protein was predicted to be a hydrophilic protein with the theoretical isoelectric point as 5.13,the instability coefficient as 35.03,and the molecular weight as 71.10 kDa.The NtHSP70-2 had high similarity with the homologous sequences of capsicum,tomato,potato and eggplant.Its secondary structure was mainly composed ofα-helix,and its tertiary structure was similar to that of CaHSP70 of capsicum.NtHSP70-2 mainly contained P-box,CGTCA-motif,ABRE,STER,MYB,DRE core and other cis-acting elements,and all them could respond to temperature stress.qRT-PCR analysis showed that the relative expression level of NtHSP70-2 gene increased in the former 12 hours after high and low temperature stresses,and then decreased gradually with the extension of stress time.NtHSP70-2 gene was expressed in roots,stems and leaves of tobacco,and the expression level was higher in leaves than that in stems and roots;the relative expression level under high temperature stress was higher than that under low temperature stress in the same part of tobacco.In summary,the NtHSP70-2 gene could respond to temperature stress and improve the tolerance of tobacco to temperature.
