摘  要:为了制备芒果苷(MGF)-莱鲍迪苷A (RebA)自组装纳米胶束(MGF-RebA SNM),并对其进行质量评价,本试验建立高效液相色谱(HPLC)方法对MGF-RebA SNM中MGF含量进行测定,考察该方法的专属性、精密度、重复性、稳定性和加样回收率;采用单因素试验考察RebA用量、 MGF用量、超声功率、超声时间和有机相比例对包封率(EE)的影响;以EE为响应值,运用Box-Behnken响应面法(RSM)优化处方;使用最优处方制备MGF-RebA SNM,测定EE、粒径、多分散指数(PDI)和Zeta电位,进行形态学观察和体外累积释放度考察,以评价其性质。结果显示,MGF含量测定方法学考察指标均良好,符合HPLC分析方法学要求;MGF-RebA SNM的最优制备工艺条件为:RebA用量11.6 mg,MGF用量12.0 mg,超声功率30%,超声时间15 min,乙醇体积34.4 mL(总体积50 mL);此条件下制备的MGF-RebA SNM样品EE为(87.14±0.56)%,粒径为(135.8±3.9)nm,PDI为0.149±0.011,Zeta电位为(-27.0±1.6)mV,外观为淡黄色澄清液体,透射电子显微镜下形态呈规则的球形,6 h累积释放度为91.43%,明显优于MGF原料药(71.40%),即胶束的形成改善了MGF的体外累计释放度。结果表明,本试验制备的MGF-RebA SNM的EE高,粒径小且均匀,体外释放度良好,可为MGF新剂型的研发提供一定的参考依据。To prepare mangiferin(MGF)-rebaudioside A(RebA)self-assembled nanomicelles(MGF-RebA SNM)and evaluate their quality,a high-performance liquid chromatography(HPLC)method was established to measure the MGF content in MGF-RebA SNM.The specificity,precision,repeatability,stability,and recovery of this method were investigated.A single-factor experiment was conducted to study the effects of RebA dosage,MGF dosage,ultrasonic power,ultrasound time,and organic phase ratio on the encapsulation efficiency(EE).Using EE as the response value,the Box-Behnken response surface method(RSM)was employed to optimize the formulation.The optimal formulation was used to prepare MGF-RebA SNM,and the EE,particle size,polydispersity index(PDI),and Zeta potential were determined.Morphological observation and in vitro cumulative release tests were conducted to evaluate its properties.The results showed that the MGF content determination method met all the required HPLC analysis criteria.The optimal preparation conditions for MGF-RebA SNM were:RebA dosage 11.6 mg,MGF dosage 12.0 mg,ultrasonic power 30%,ultrasound time 15 min,and ethanol volume 34.4 mL(total volume 50 mL).The MGF-RebA SNM samples prepared under these conditions had an EE of(87.14±0.56)%,a particle size of(135.8±3.9)nm,PDI of 0.149±0.011,and Zeta potential of(-27.0±1.6)mV.The appearance was a pale yellow clear liquid,and the morphology observed under transmission electron microscopy was a regular spherical shape.The 6-hour cumulative release rate was 91.43%,significantly better than the MGF raw material(71.40%),indicating that the formation of the micelles improved the in vitro cumulative release of MGF.The results suggest that the prepared MGF-RebA SNM have a high EE,small and uniform particle size,and good in vitro release,providing a useful reference for the development of new MGF formulations.
