血根碱联合β-内酰胺类抗生素对金黄色葡萄球菌的抗菌作用

摘  要:为了解奶牛乳房炎源金黄色葡萄球菌(S.aureus)对临床常用β-内酰胺类抗生素的耐药情况,及血根碱(SAN)与β-内酰胺类抗生素对耐药S.aureus的协同抑菌作用,本试验从临床型奶牛乳房炎样本中分离得到57株S.aureus,采用微量肉汤稀释法检测8种临床常用β-内酰胺类抗生素对其的最小抑菌浓度(MIC);采用微量肉汤棋盘稀释法检测SAN与β-内酰胺类抗生素的联合抑菌指数(FICI),通过体外时间-杀菌曲线检测SAN协同β-内酰胺类抗生素对S.aureus的杀伤作用。结果显示,S.aureus对β-内酰胺类抗生素的耐药率为59.65%,其中对氨苄西林(40.35%)、青霉素(35.09%)和阿莫西林(35.09%)严重耐药;SAN对青霉素、氨苄西林、阿莫西林和头孢氨苄均有协同作用,可降低β-内酰胺类抗生素MIC 2~64倍;时间-杀菌曲线显示,SAN可以增强青霉素、氨苄西林、阿莫西林和头孢氨苄对耐药S.aureus的杀伤作用。结果表明,SAN能够抑制耐药细菌的生长,恢复β-内酰胺类抗生素对S.aureus的抗菌活性,为S.aureus导致的奶牛乳房炎的治疗提供科学参考。To investigate the antibiotic resistance of Staphylococcus aureus(S.aureus)isolated from bovine mastitis against commonly usedβ-lactam antibiotics,and to explore the synergistic antibacterial effects of sanguinarine(SAN)combined withβ-lactam antibiotics on drug-resistant S.aureus,57 strains of S.aureus were isolated from clinical bovine mastitis samples.The minimal inhibitory concentration(MIC)of eight commonly usedβ-lactam antibiotics was determined by the microdilution broth method.The fractional inhibitory concentration indices(FICI)of SAN andβ-lactam antibiotics was determined using the microdilution checkerboard dilution method,and the bactericidal effect of SAN combined withβ-lactam antibiotics was tested using the in vitro time-kill curve.The results showed that the resistance rate of S.aureus toβ-lactam antibiotics was 59.65%,with severe resistance to ampicillin(40.35%),penicillin G(35.09%),and amoxicillin(35.09%).SAN exhibited synergistic effects with penicillin G,ampicillin,amoxicillin,and cefalexin,reducing the MIC ofβ-lactam antibiotics by 2-64 times.The time-kill curves indicated that SAN could enhance the bactericidal effect of penicillin G,ampicillin,amoxicillin,and cefalexin on drug-resistant S.aureus.The results suggest that SAN can inhibit the growth of resistant bacteria and restore the antibacterial activity ofβ-lactam antibiotics against S.aureus,providing scientific reference for the treatment of bovine mastitis caused by S.aureus.
