摘 要:为了揭示包载聚乳酸-羟基乙酸共聚物(PLGA)的蒲公英多糖纳米粒(PLGA-DP-NP)对小鼠单核巨噬细胞白血病细胞(Raw264.7细胞)免疫效果的影响,本试验以Raw264.7细胞为研究对象,采用细胞增殖/毒性检测(CCK-8)法测定PLGA-DP-NP对Raw264.7细胞的毒性和细胞增殖的影响,用酶联免疫吸附试验(ELISA)测定PLGA-DP-NP对Raw264.7细胞分泌的细胞因子白介素-2(IL-2)和白介素-6(IL-6)的影响,采用流式细胞术检测低、中、高浓度(18.8、75、150μg/mL)PLGA-DP-NP对Raw264.7细胞成熟标志分子CD80和CD86表达,以及对Raw264.7细胞吞噬卵清蛋白(OVA)的影响。结果显示,PLGA-DP-NP浓度低于500μg/mL时,Raw264.7细胞的存活率较高;与阴性对照组相比,18.8、37.5、75、150和300μg/mL的PLGA-DP-NP均可显著促进细胞增殖(P <0.05),均能显著增加Raw264.7细胞的IL-2和IL-6分泌量(P <0.05);低、中、高浓度的PLGA-DP-NP均能增强Raw264.7细胞CD80(P> 0.05或P <0.01或P <0.05)和CD86(P <0.01)的表达,提高Raw264.7细胞吞噬OVA百分比(P <0.01),浓度为75μg/mL时增幅最大。结果表明,安全浓度范围内的PLGA-DP-NP可以显著促进Raw264.7细胞增殖,提高细胞因子IL-2和IL-6分泌,促进细胞成熟,提升细胞的吞噬作用,浓度为75μg/mL时效果最佳。To investigate the immune effects of poly(lactic-co-glycolic acid)(PLGA)-encapsulated Dandelion polysaccharide nanoparticles(PLGA-DP-NP)on RAW264.7 murine macrophage cells,this study used RAW264.7 cells as the research model.The cytotoxicity and proliferation effects of PLGA-DP-NP on RAW264.7 cells were assessed using the CCK-8 assay.The secretion levels of interleukin-2(IL-2)and interleukin-6(IL-6)were measured using an enzyme-linked immunosorbent assay(ELISA).Flow cytometry was employed to examine the expression of the cell maturation markers CD80 and CD86 and the phagocytosis of ovalbumin(OVA)by RAW264.7 cells under low,medium,and high concentrations(18.8,75,and 150μg/mL)of PLGA-DP-NP.The results showed that RAW264.7 cell viability remained high when the PLGA-DP-NP concentration was below 500μg/mL.Compared with the Negative control group,PLGA-DP-NP at concentrations of 18.8,37.5,75,150,and 300μg/mL significantly promoted cell proliferation(P<0.05)and increased the secretion of IL-2 and IL-6(P<0.05).Low,medium,and high concentrations of PLGA-DP-NP enhanced the expression of CD80(P>0.05,P<0.01,or P<0.05)and CD86(P<0.01)in RAW264.7 cells,and increased the percentage of Raw264.7 cells that phagocytose OVA(P<0.01),with the most pronounced effect observed at 75μg/mL.These findings indicate that PLGA-DP-NP,within a safe concentration range,can significantly promote RAW264.7 cell proliferation,enhance IL-2 and IL-6 secretion,facilitate cell maturation,and improve phagocytic activity,with the optimal effect at 75μg/mL.
