摘  要:目的:建立一测多评法(QAMS)同时测定加味通幽汤中5种成分的含量,为加味通幽汤的质量控制、临床研究等提供理论依据。方法:采用Diamonsil C_(18)色谱柱(4.6 mm×250 mm,5μm),以乙腈-0.05%甲酸水溶液为流动相,325 nm为检测波长进行梯度洗脱。以异阿魏酸为参照物,计算其与阿魏酸、野黄芩苷、羟基红花黄色素A、槲皮素的相对校正因子,利用外标法(ESM)与QAMS分别计算5种成分的含量。结果:在一定线性范围内,异阿魏酸相对阿魏酸、野黄芩苷、羟基红花黄色素A、槲皮素的相对校正因子重复性良好,ESM与QAMS计算所得的5种成分的含量无明显差异。结论:该实验所建立的QAMS法简便稳定、重复性好、专属性强,可用于加味通幽汤的质量控制。Objective:To develop a quantitative analysis of multi-components by a single marker(QAMS)method for the simultaneous determination of the content of 5 ingredients in modified tongyou decoction,providing theoretical basis for quality control and clinical research of modified tongyou decoction.Methods:Diamonsil C_(18) column(4.6 mm×250 mm,5μm)was used for gradient elution with acetonitrile-0.05% formic acid aqueous solution as mobile phase and 325 nm as detection wavelength.The relative correction factors of ferulic acid,scutellarin,hydroxysafflor yellow A,and quercetin were calculated,using the external standard method(ESM)and QAMS to calculate the contents of 5 components,respectively.Results:In a certain linear range,the repeatability of the relative correction factors of isoferulic acid to ferulic acid,scutellarin,hydroxysafflor yellow A,and quercetin was good,and there was no significant difference in using the external standard method(ESM)and QAMS to calculate the contents of 5 components,respectively.Conclusion:The QAMS method established in this experiment is simple,stable,reproducible,and specific,and can be used for quality control of modified tongyou decoction.
