摘 要:目的:分析转化生长因子β1(TGF-β1)诱导的调节性T细胞抑制炎性骨缺损吸收的修复机制。方法:选择25只健康新西兰大白兔构建高特异性兔膝关节股骨髁骨缺损动物模型,最终成功构建20只动物模型,随机分为A组和B组,每组各10只,同时纳入10只健康新西兰大白兔作为空白对照组。A组于双侧膝关节皮下注射5 ng/mL的TGF-β1溶液(2 d/次),空白对照组注射等体积生理盐水,B组不注射,三组均常规饲养14 d。比较三组的血清炎性因子水平、膝关节股骨髁骨缺损组织中微生物DNA甲基转移酶1(DNMT1)、TGF-β1、Runt相关转录因子2(Runx2)和骨桥蛋白(OPN)表达等情况。结果:B组血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、C反应蛋白(CRP)水平均高于A组和空白对照组,差异有统计学意义(P<0.05);A组和空白对照组血清TNF-α、IL-6、CRP水平比较,差异无统计学意义(P>0.05)。B组膝关节股骨髁骨缺损组织中DNMT1、TGF-β1、Runx2和OPN表达低于A组和空白对照组,且A组低于空白对照组,差异有统计学意义(P<0.05)。结论:TGF-β1可减轻高特异性兔膝关节股骨髁骨缺损的炎症反应,促进成骨分化。Objective:To analyze the repair mechanism of Transforming Growth Factor-betal(TGF-β1)induced by regulatory T cells to inhibiting the absorption of inflammatory bone defect.Method:A total of 25 healthy New Zealand white rabbits were selected to construct a high-specificity animal model of femoral condylar bone defect of knee joint.Finally,20 animal models were successfully constructed and randomly divided into group A and group B with 10 rabbits each,and 10 healthy New Zealand white rabbits were included as a blank control group.The group A were injected subcutaneously with 5 ng/mL TGF-β1 solution(2 days/time)on both knee joints,the blank control group were injected with equal volume of normal saline,group B were not injected,and rabbits in the three groups were routinely fed for 14 days.The levels of serum inflammatory factors and the expressions of microbial DNA methyltransferase 1(DNMT1),TGF-β1,RUNt-related transcription factor 2(Runx2)and osteopontin(OPN)in femoral condylar bone defect of knee joint of rabbits were compared among the three groups.Result:The serum levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and C-reactive protein(CRP)in group B were higher than those in group A and blank control group,and the differences were statistically significant(P<0.05).There was no significant difference in serum TNF-α,IL-6 and CRP levels between group A and blank control group(P>0.05).The expressions of DNMT1,TGF-β1,Runx2 and OPN in femoral condylar bone defect tissue of knee joint of rabbits in group B were lower than those in group A and blank control group,and the differences were statistically significant(P<0.05).Conclusion:TGF-β1 can alleviate the inflammatory response of high specificity rabbit knee joint femoral condylar bone defects and promote osteogenic differentiation.
