摘 要:策勒黑羊是多胎多产于一体的绵羊品种,具有常年发情和繁殖率高的特征,以优良的多胎性状闻名中外。而绵羊多胎性状的QTLs(Quantitative Trait Locus)多存在于OAR6(Ovis aries 6绵羊6号染色体)上,文章通过连锁不平衡分析即LD分析,解析该染色体上分子标记之间的遗传性。对策勒黑羊采集耳组织,提取DNA,制备绵羊基因组芯片,获得数据;从国际基因组数据库下载获得引入绵羊品种芯片数据。利用PLINK软件进行数据整合、质量控制和LD计算,获得强连锁区域,并进行基因注释,得到51个基因,分析整理后发现这些基因与机体生理调控、致癌和激素调节等有关。试验对基因进行筛选,挑出与多胎性状等有关的GNRHR、RPL21、BMP3基因,是影响繁殖性状的候选基因。The Qira black sheep,renowned globally for its outstanding polytocous trait,is a sheep breed that integrates multiplicity trait and high fertility,characterized by perennial estrus and high reproductive rates.The quantitative trait loci(QTLs)associated with the polytocous trait in sheep are predominantly located on Ovine Chromosome 6(OAR6).To decipher the genetic relationships among molecular markers on this chromosome,linkage disequilibrium(LD)analysis was conducted.Ear tissues were collected from Qira black sheep for DNA extraction,followed by the preparation of sheep genomic chips to acquire data.Additionally,sheep breed chip data were obtained from international genomic databases.Utilizing PLINK software,the data were integrated,the quality control was conducted,and LD calculations were performed,resulting in the identification of strong linkage regions.Subsequently,gene annotation revealed 51 genes associated with these regions,which upon further analyses were found to be related to physiological regulation,carcinogenesis,and hormone modulation in the organism.Through rigorous screening,GNRHR,RPL21 and BMP3 were singled out as candidate genes significantly associated with the polytocous trait and reproductive performance.
