摘 要:目的研究小檗碱(BBr)对脂多糖(LPS)诱导的胸主动脉环损伤的影响,并初步探讨其作用机制。方法快速分离昆明小鼠胸主动脉并固定于多通道血管张力测试系统中,以乙酰胆碱(Ach)或硝普钠(SNP)检测内皮或非内皮依赖的血管功能,使用不同浓度BBr(0.1~10μmol/L)刺激离体胸主动脉环30min,然后用1μg/mL LPS处理2h。记录LPS诱导的血管张力变化情况,使用试剂盒检测胸主动脉匀浆中的一氧化氮(NO)和丙二醛(MDA)水平变化情况。用左旋硝基精氨酸甲酯(L-NAME)1mmol/L或Tempol 1mmol/L预孵育小鼠胸主动脉环30min,重复上述药物处理步骤,记录血管张力的变化。结果LPS能显著降低NO含量,增加MDA含量,LPS使Ach诱导的内皮依赖性舒张作用受损。BBr处理后,呈剂量依赖性缓解上述情况。L-NAME可以阻断BBr的此种作用,Tempol对BBr的作用无影响。然而,SNP诱导的非内皮依赖性舒张作用在所有组中都没有改变。结论BBr通过调节NO信号,对LPS诱导的主动脉内皮功能障碍的氧化应激损伤具有保护作用。Objective To investigate the effect and mechanism of berberine(BBr)on lipopolysaccharide(LPS)-induced impairment of thoracic aortic rings.Methods The Kunming mice thoracic aortic rings were quickly isolated and suspended in a Multi Myograph System.The isolated rings were pretreated with different concentration of BBR(0.1~10μmol/L)for 30 min,and then treated with 1μg/mL LPS for 2 hours.The changes of isometric tension were recorded induced by LPS.The Nitric Oxide(NO)and malondialdehyde(MDA)levels of thoracic aorta ring homogenate were measured by the kits.Aortic rings isolated from mice were preincubated with L-nitroarginine methyl methyl(L-NAME)1 mmol/L or Tempol 1 mmol/L for 30 minutes.The above drug treatment steps were repeated,as well as the record of isometric tension.Results LPS dramatically decreased the content of NO and increased the level of MDA.LPS dramatically impaired acetyl choline(Ach)-induced endothelium-dependent relaxation(all P<0.05).After the treatment of BBr,the above situation was alleviated in a dose-dependent manner.L-NAME blocked the function of BBr.Tempol had no effect on the function of BBr.However,sodium nitroprusside(SNP)-induced endothelium-independent relaxation was not altered in all groups.Conclusion BBr plays a protective role in LPS-induced oxidative stress injury of endothelial dysfunction in aortic arteries isolated from Kunming mice ex vivo by regulating NO signaling.
