基于宏基因组学的慢性心衰心气阴虚证大鼠肠道微生物改变的研究

摘  要:目的应用宏基因组学技术探讨心气阴虚证心力衰竭对肠道微生物的特异性影响。方法运用随机数字表法对18只大鼠(Dahl盐敏感大鼠12只,耐盐大鼠6只)编号,将Dahl盐敏感大鼠随机均分为对照组(CON,接受0.3%NaCl低盐饮食)和模型组(H-HF,接受8%NaCl高盐饮食),耐盐组(SR)大鼠同样接受高盐饮食,持续喂养20周。干预结束后,收集新鲜粪便,运用宏基因组学技术比较三组大鼠肠道微生物结构和基因功能差异。结果Venn图结果显示,CON组与SR组共有的基因数目最多,其次是H-HF组与SR组,CON组和H-HF组共有的基因数目最少;PCoA图结果显示,CON组样本与SR组样本相对聚集,H-HF组样本与CON组、SR组样本相对离散;LEfSe结果显示,与CON组比较,H-HF组有益菌丰度显著降低,机会致病菌丰度显著升高;CAZy数据库、GO数据库、KEGG数据库注释结果表明,与碳水化合物代谢、脂类代谢、氨基酸代谢等相关的基因水平在H-HF组大鼠体内异常表达。结论肠道微生物的特异性改变与心气阴虚证心力衰竭的病理过程密切相关。Objective To investigate the specific impact of heart failure with qi and yin deficiency syndrome on the gut microbiota using metagenomic techniques.Methods Eighteen rats(12 Dahl salt-sensitive rats and 6 salt-resistant rats)were numbered using a random number table method.Dahl salt-sensitive rats were randomly divided into a control group(CON,receiving a low salt diet of 0.3%NaCl)and a model group(H-HF,receiving a high salt diet of 8%NaCl).Salt-resistant(SR)rats also received a high salt diet,and all rats were fed for 20 weeks.After the intervention,fresh fecal samples were collected,and metagenomic techniques were employed to compare the structural and functional differences in the gut microbiota among the three groups of rats.Results Venn diagram revealed the highest number of shared genes between the CON and SR groups,followed by the H-HF and SR groups,with the least number of shared genes between the CON and H-HF groups.The results of PCoA diagram showed that samples from the CON group clustered relatively closely with those from the SR group,while samples from the H-HF group were relatively discrete from both the CON and SR groups.LEfSe analysis indicated a significant reduction in the abundance of beneficial bacteria and a marked increase in the abundance of opportunistic pathogens in the H-HF group compared to the CON group.Annotations from the CAZy,GO,and KEGG databases suggested abnormal expression of genes related to carbohydrate metabolism,lipid metabolism,and amino acid metabolism in the H-HF group rats.Conclusion Investigating the gut microbiota provides a novel perspective for understanding the essence of traditional Chinese medicine syndromes in heart failure.
