摘  要:为系统分析PPP1R3C所在基因家族于牛及其他物种间的进化水平、分子结构及功能特性,本研究利用生物信息学技术分析了PPP1R3C所属家族—PPP1R3基因家族基因特征,并采用RT-qPCR检测PPP1R3C基因的表达水平。结果表明:1)PPP1R3基因家族成员在等电点和分子量上存在显著差异。2)进化树和保守结构域分析表明,PPP1R3基因家族成员在进化过程中形成了不同的基序和结构域组合;共线性分析表明了PPP1R3基因家族成员在牛与小鼠、人类、绵羊及家猪染色体之间的位置变化规律。3)RT-qPCR检测结果显示,PPP1R3C基因在牛的脂肪和肺组织中具有显著的高表达,而在肌肉相关组织中的表达水平最低;随着牛脂肪细胞分化成熟,其表达水平逐步提升。综上,本研究探究了PPP1R3基因家族在牛及其他物种间的进化水平、分子结构及功能特性,揭示了PPP1R3C基因在脂肪代谢和能量平衡中的关键调控作用,为深入研究该基因家族在不同生理和病理状态下的具体机制提供了一定的理论基础。To systematically analyze the evolutionary relationships,molecular structures,and functional characteristics of the PPP1R3C gene family across bovine and other species,this study employed bioinformatics approaches to investigate the genetic features of the PPP1R3 gene family,to which PPP1R3C belongs,and examined PPP1R3C expression levels using RT-qPCR.The results revealed:1)Members of the PPP1R3 gene family exhibited significant variations in their isoelectric points and molecular weights.2)Phylogenetic tree and conserved domain analyses demonstrated that PPP1R3 family members have evolved distinct motif and domain combinations throughout their evolution;synteny analysis revealed patterns of positional changes of PPP1R3 family members across the chromosomes of cattle,mice,humans,sheep,and pigs.3)RT-qPCR analysis demonstrated that PPP1R3C exhibits significantly high expression in bovine adipose and lung tissues,while showing minimal expression in muscle-related tissues;furthermore,its expression levels progressively increased during bovine adipocyte differentiation and maturation.In conclusion,this study investigated the evolutionary relationships,molecular structures,and functional characteristics of the PPP1R3 gene family across bovine and other species,highlighting the crucial regulatory role of PPP1R3C in lipid metabolism and energy homeostasis.These findings provide a theoretical foundation for further investigation of the specific mechanisms of this gene family under various physiological and pathological conditions.
