摘  要:为探究circFAM53B在奶牛乳房炎中的功能,以牛乳腺上皮细胞(bMECs)为研究对象,采用生物信息学对circFAM53B的功能进行预测,然后在LPS诱导的bMECs炎症中干扰或过表达circFAM53B检测其对细胞炎症、增殖和凋亡的影响。结果表明:1)奶牛circFAM53B为真实存在的环状RNA,主要定位于bMECs细胞质中。2)在LPS诱导bMECs炎症反应中,circFAM53B的表达水平极显著上调(P<0.01)。3)在bMECs炎症反应中,奶牛circFAM53B可促进炎症因子的表达和分泌,抑制细胞活力和增殖,促进细胞凋亡,加剧炎症的发展。综上,circFAM53B可能参与奶牛乳房炎的调控,促进炎症进程。本研究为奶牛乳房炎抗病分子育种奠定基础。To investigate the function of circFAM53B in dairy cow mastitis,this study took bovine mammary epithelial cells(bMECs)as the research object,and bioinformatics was used to predict the function of circFAM53B.The effects of circFAM53B on cell inflammation,proliferation and apoptosis were examined by interfering or overexpressing circFAM53B in LPS-induced bMECs inflammation.The results showed that:1)Dairy cow circFAM53B was a real-existing circular RNA,mainly located in the cytoplasm of bMECs.2)The expression level of dairy cow circFAM53B was highly significantly up-regulated in LPS-induced inflammatory response of bMECs(P<0.01).3)In the inflammatory response of bMECs,circFAM53B in cows can promote the expression and secretion of inflammatory factors,inhibits cell viability and proliferation,and promote apoptosis in bMECs inflammation,and exacerbate the development of inflammation.In conclusion,circFAM53B may be involved in the regulation of cow mastitis and promote the inflammatory process.This study lays a foundation for molecular breeding for resistance to cow mastitis.
