摘  要:为探究富集在mTOR通路上GRB10、ULK2和PIK3R1基因在鸡脂肪沉积和肌肉发育中的调控作用,本研究以S3、F和H系为试验素材,分析其屠宰性能、胸肌和血清中甘油三酯(TG)、磷脂(PL)、胆固醇(CHO)含量差异,再利用荧光定量PCR技术检测3个基因在不同组织及细胞中的表达变化,并做相关性分析。结果表明:1)1)F系在体重、全净膛重、胸肌重、腿肌重、腹脂重显著最高(P<0.05),H系腹脂率显著最低(P<0.05)。2)不同发育时期胸肌和血清中的脂质含量均呈波浪型变化规律。胸肌中TG含量在2和15周龄时S3系显著最高(P<0.05),CHO在2和5周龄时H系显著最低(P<0.05);血清中TG含量在5周龄时F系最高(P<0.05),LDL-C(低密度脂蛋白)在2、5和15周龄时F系显著最高(P<0.05)。3)H系肌纤维直径/密度显著低于/高于其它品系(P<0.05),肌纤维的横截面积则相反,S3系脂肪细胞的体积显著最大(P<0.05)。4)3个基因在不同品系鸡不同周龄的肝脏、腹脂、胸肌、腿肌、皮脂组织、成肌细胞和肌内脂肪细胞中均有表达,呈现出显著的品系和组织差异性(P<0.05)。5)GRB10、ULK2和PIK3R1表达与胸肌重(率)、腿肌重(率)呈显著正相关(P<0.05),ULK2与腹脂重(率)显著正相关(P<0.05)。综上,不同品种的鸡在成长发育过程中,胸肌和血清中的脂质水平波动明显;3个基因在多个组织中表达丰富,且在品种和组织间有显著差异,并与胸肌重(率)、腿肌重(率)、腹脂重(率)呈显著相关,是研究鸡肌肉发育和脂脂沉积的潜在靶点。本研究为深入分析GRB10、ULK2和PIK3R1的功能和机制提供了坚实的理论基础和数据支撑。In order to investigate the regulatory role of GRB10,ULK2 and PIK3R1 genes enriched in the mTOR pathway in chicken fat deposition and muscle development,the present study was conducted to analyse the differences in slaughter performance,pectoral muscle and serum levels of triglycerides(TG),phospholipids(PL),and cholesterol(CHO)using the S3,F,and H lines,and then to detect the expression changes of the three genes using fluorescence qPCR in expression changes in different tissues and cells,and do correlation analysis.The results showed that:1)The F line had the significantly highest(P<0.05)in body weight,full net chamber weight,pectoral muscle weight,leg muscle weight and abdominal fat weight,and the H line had the significantly lowest(P<0.05)in abdominal fat rate.2)Lipid contents in pectoral muscle and serum showed a wave pattern of change in different developmental periods.The TG content in the pectoral muscle was significantly(P<0.05)highest in the S3 line at 2 and 15 weeks of age,and CHO was significantly(P<0.05)lowest in the H line at 2 and 5 weeks of age;the TG content in the serum was significantly(P<0.05)highest in the F line at 5 weeks of age,and the LDL-C was significantly(P<0.05)highest in the F line at 2,5,and 15 weeks of age.3)The diameter/density of muscle fibres in the H line was significantly lower/higher than that of the other lines.density was significantly lower/higher than that of other lines(P<0.05),while the cross-sectional area of myofibrils was the opposite,with adipocytes in line S3 having the significantly largest volume(P<0.05).4)The three genes were expressed in liver,abdominal lipids,pectoral muscles,leg muscles,sebaceous tissues,adult and intramuscular myofibroblasts in chickens of different lines at different weekly ages,showing significant line and tissue variability(P<0.05).5)GRB10,ULK2 and PIK3R1 expression showed significant positive correlation with pectoral muscle weight(rate)and leg muscle weight(rate)(P<0.05),and ULK2 was significantly positively correlated with abdominal fat
