摘 要:采用高效液相色谱法建立10批次不同产地马蹄金指纹图谱,并进行聚类分析。称取0.5 g马蹄金,以70%乙醇为溶剂,加热回流提取样品溶液,使用CACELLPAK-C_(18)色谱柱,流动相为乙腈-0.2%(体积比)磷酸水溶液,梯度洗脱,流量为1.0 m L/min,检测器波长为254 nm,建立马蹄金指纹图谱。10批次马蹄金相似度为0.97~0.99,相对保留时间测定结果的相对标准偏差(RSD)小于1.0%。通过共有模式筛选出8个共有特征峰,其中指认主要1个共有峰为芦丁。方法的重复性、稳定性测试的色谱峰面积RSD均小于1.0%,芦丁的样品加标回收率为98.78%~101.56%,RSD为1.0%(n=6),符合指纹图谱检测要求。运用SPSS 19.0软件进行聚类分析,马蹄金样品分为两大类。该方法具有良好的重复性和稳定性,结合相似度评价和聚类分析,可以评价不同产地马蹄金质量,为马蹄金的质量控制提供参考及对其产地的初步鉴别。The fingerprints of 10 batches of Dichondra repens from different habitats were established by high performance liquid chromatography,and cluster analysis was carried out.A total of 0.5 g of Dichondra repens was weighed,and the sample solution was extracted by heating reflux with 70%ethanol.The chromatographic column was CACELL PAK-C_(18),and the mobile phase was acetonitrile-0.2%(volume ratio)phosphoric acid aqueous solution with gradient elution.The flow rate was 1.0 mL/min,and the detector wavelength was 254 nm to establish the fingerprint of Dichondra repens.The similarities of 10 batches of dichondra were between 0.97 to 0.99,and the relative standard deviation of relative retention time test results was less than 1.0%.Eight common characteristic peaks were screened out by the common mode,and one of the main common peaks was identified as rutin.The RSD of the chromatographic peak area of the repeatability and stability test of the method was less than 1.0%.The recovery rate of rutin was 98.78%-101.56%,and the RSD was 1.0%(n=6),which met the requirements of fingerprint detection.Cluster analysis was performed by using SPSS 19.0 software,and the samples were divided into two categories.The method has good repeatability and stability.Combined with similarity evaluation and cluster analysis,the quality of Dichondra repens from different habitats can be evaluated,which can provide reference for the quality control of Dichondra repens and the preliminary identification of its origin.
