消痰化瘀利窍方抑制细胞焦亡减轻CIH小鼠胸主动脉损伤的研究

摘  要:目的:探讨消痰化瘀利窍方(XHLF)对慢性间歇性低氧(Chronic Intermittent Hypoxia, CIH)小鼠胸主动脉细胞焦亡的抑制作用及可能机制。方法:将24只健康的8周龄雄性C57BL/6N小鼠随机分成为常氧对照组(Con组)、慢性间歇性低氧暴露组(CIH组)、慢性间歇性低氧+消痰化瘀利窍方组(CIH+XHLF组)、常氧+消痰化瘀利窍方组(Con+XHLF组)。CIH组与CIH+XHLF组暴露于CIH的环境中,Con+XHLF组和CIH+XHLF组每日给予消痰化瘀利窍方灌胃[26.8 g/(kg·d)],而CIH组与Con组给予同等体积生理盐水,共计干预28 d。造模结束后采用尾套法监测小鼠血压;HE染色观察胸主动脉病理改变;检测血清中内皮素-1(ET-1)和一氧化氮(NO)含量;DHE染色观察活性氧(ROS)水平,比色法检测胸主动脉中丙二醛(MDA)含量、总超氧化物歧化酶(T-SOD)活力;观察胸主动脉TUNEL染色阳性细胞率;免疫组织化学染色检测NOD样受体蛋白3(NLRP3)、胱天蛋白酶-1(Caspase-1)、白细胞介素-1β(IL-1β)蛋白表达。结果:与Con组相比,CIH组小鼠收缩压升高(P<0.05);血管内膜不平整,细胞核排列紊乱;ET-1含量升高,NO含量下降(P<0.05);ROS、MDA水平增加;T-SOD活力下降(P<0.05);主动脉TUNEL染色阳性细胞率增加,NLRP3、Caspase-1和IL-1β蛋白表达增强(P<0.05)。与CIH组相比,CIH+XHLF组小鼠血压下降(P<0.05);血管内皮损伤减轻,ET-1含量降低,NO含量增高(P<0.05);ROS、MDA水平下降;T-SOD活力上升(P<0.05);主动脉TUNEL染色阳性细胞率下降,NLRP3、Caspase-1、IL-1β蛋白表达下降(P<0.05)。结论:XHLF可能通过抑制ROS/NLRP3/Caspase-1通路减轻CIH小鼠胸主动脉损伤。Objective:To explore the inhibitory effect of Xiaotan Huayu Liqiao Formula(XHLF)on pyroptosis of thoracic aortic cells in mice with Chronic Intermittent Hypoxia(CIH)and its possible mechanism.Methods:Twenty-four healthy 8-week-old male C57BL/6N mice were randomly divided into a normoxic control group(Con group),a chronic intermittent hypoxia exposure group(CIH group),a chronic intermittent hypoxia+Xiaotan Huayu Liqiao Formula group(CIH+XHLF group),and a normoxic+Xiaotan Huayu Liqiao Formula group(Con+XHLF group).The CIH group and CIH+XHLF group were exposed to a CIH environment,and the Con+XHLF group and CIH+XHLF group were given Xiaotan Huayu Liqiao Formula by gavage daily,while the CIH group and Con group were given an equal volume of physiological saline,with a total intervention period of 28 days.After the modeling was completed,the blood pressure of the mice was monitored using the tail-cuff method;HE staining was used to observe the pathological changes in the thoracic aorta;the levels of endothelin-1(ET-1)and nitric oxide(NO)in the serum were detected;DHE staining was used to observe the levels of reactive oxygen species(ROS),and colorimetric method to detect the content of malondialdehyde(MDA)and the activity of total superoxide dismutase(T-SOD)in the thoracic aorta;the positive cell rate of TUNEL staining in the thoracic aorta was observed;immunohistochemical staining was used to detect the protein expression of NOD-like receptor protein 3(NLRP3),Caspase-1,and interleukin-1β(IL-1β).Results:Compared with the Con group,the systolic blood pressure of the CIH group was significantly increased(P<0.05);the vascular intima was uneven,and the arrangement of cell nuclei was disordered;the content of ET-1 increased,and NO decreased(P<0.05);the levels of ROS and MDA increased;the activity of T-SOD decreased(P<0.05);the positive cell rate of TUNEL staining in the aorta increased,and the protein expression of NLRP3,Caspase-1,and IL-1βwas enhanced(P<0.05).Compared with the CIH group,the blood pressure of the CIH+XH
