摘 要:目的比较薄层液基细胞技术制片仪器法与自然沉降法在制片质量和效果上的差异,评估自然沉降法作为仪器法的应急预案的可行性。方法收集南京金域医学检验有限公司病理科已发布诊断报告的39例宫颈液基细胞学检查剩余原始样本,分别采用薄层液基细胞技术的仪器法与自然沉降法进行制片,并进行免疫细胞化学双染(P16/Ki-67)。结果自然沉降法制备的玻片染色质量良好,背景清晰,细胞平铺均匀。在沉降时长为30~50 min时,能够富集颈管上皮细胞数量(28.8~76.9个),细胞重叠率为3%~10%,其效果与仪器法相似。双染试验证明,经过自然沉降30~50 min后,可富集足够的阳性细胞用于辅助诊断。结论薄层液基细胞技术自然沉降法的制片质量和诊断结果与仪器法一致,能够作为薄层液基细胞学仪器法制片的应急预案,具有可行性,并可广泛应用于宫颈细胞学制片工作中。Objective To compare the differences in slide preparation quality and effectiveness between the instrument-based method and the natural sedimentation method of thin-layer liquid-based cytology,and to evaluate the feasibility of using the natural sedimentation method as an emergency backup for the instrument-based method.Methods 39 cervical liquid-based cytology samples with published diagnostic reports from the Pathology Department of Nanjing KingMed Diagnostics were collected.These samples were prepared using both the instrument-based method and the natural sedimentation method,followed by immunocytochemical double staining(P16/Ki-67).Results Slides prepared by the natural sedimentation method showed satisfactory staining quality,clear background,and even cell distribution.When the sedimentation duration was between 30 to 50 minutes,the number of cervical epithelial cells enriched ranged from 28.8 to 76.9 cells,with a cell overlap rate of 3%to 10%.The effect was similar to that of the instrument-based method.Double staining tests demonstrated that after 30 to 50 minutes of natural sedimentation,sufficient positive cells could be enriched for auxiliary diagnosis.Conclusion The slide preparation quality and diagnostic results of the natural sedimentation method in thin-layer liquid-based cytology are consistent with those of the instrument-based method.It can serve as a feasible emergency backup for the instrument-based method and has the potential for broad application in cervical cytology slide preparation.
