摘 要:为了进一步研究绒山羊次级毛囊干细胞(SHFSCs)的功能,试验采用调整电压和脉冲时间的方法对绒山羊SHFSCs的电转染条件进行了研究,首先采用不同的电压条件对绒山羊SHFSCs进行电转染,对筛选得到的较优电压条件进行脉冲时间优化,以期提高绒山羊SHFSCs电转染效率。结果表明:电压偏低时转染效率过低,电压过高和脉冲时间过长时均会导致SHFSCs大量死亡。绒山羊SHFSCs最佳的电转染条件为275 V/5 ms,在该参数下约有49.97%的细胞被成功转染,细胞存活率较高且形态较好。说明通过调整电压和脉冲时间可提高SHFSCs电转染效率。In order to further explore the function of cashmere goat secondary hair follicle stem cells,the method of adjusting voltage and pulse time was used to study the electrotransfection conditions of cashmere goat SHFSCs.First,different voltage conditions were used for electrotransfection of hair follicle stem cells,and then the pulse time of the better voltage was optimized to improve the transfection efficiency of cashmere goat SHFSCs.The results showed that the transfection efficiency was too low at low voltage,and the high voltage and long pulse duration would lead to the death of SHFSCs.The optimal electrotransfection condition for cashmere goat hair follicle stem cells was 275 V/5 ms,and about 49.97%of the cells were successfully transfected under this parameter,with high cell survival rate and better morphology.The results indicated that the transfection efficiency of cashmere goat SHFSCs could be improved by adjusting the voltage and pulse time.
