摘 要:为了解河北省部分地区太行鸡群中流行的禽白血病病毒(Avian leukosis virus,ALV)亚群及其遗传进化特征,试验于2022年7月份—2023年3月份采集河北省保定市、石家庄市10个养殖场太行鸡肝脏和脾脏组织的混合样本97份,采用RT-PCR方法分别检测样本中ALV-A、ALV-B、ALV-J、ALV-K,并采用DNASTAR、MegAlign等分子生物学软件对检出的部分毒株(随机选取)的env基因测序结果进行遗传进化分析。结果表明:组织病料样本中,ALV总检出率为60.8%(59/97);其中ALV-A、ALV-B检出率均为0(0/97),ALV-J检出率为19.6%(19/97),ALV-K检出率为55.7%(54/97),ALV-J与ALV-K的混合检出率为14.4%(14/97)。选取的9株ALV-J env基因之间的核苷酸和氨基酸相似性分别为92.5%~100%和88.1%~100%。选取的9株ALV-J与15株ALV参考毒株env基因的核苷酸和氨基酸相似性分别为59.7%~97.6%和51.0%~97.4%;其中与ALV-J参考毒株env基因的核苷酸和氨基酸的相似性较高,分别为84.0%~97.6%和84.9%~97.4%;与ALV-J原型株HPRS103的核苷酸和氨基酸相似性分别为93.0%~95.4%和88.8%~94.7%;与ALV-A、ALV-B、ALV-C、ALV-D、ALV-E、ALV-K参考毒株的核苷酸和氨基酸相似性较低。与ALV-J原型株HPRS103相比,选取的9株ALV-J存在28处突变位点和1处缺失位点。选取的20株ALV-K env基因之间的核苷酸和氨基酸相似性分别为96.0%~99.7%和94.0%~99.2%;选取的20株ALV-K与11株ALV参考毒株env基因的核苷酸和氨基酸相似性分别为51.7%~99.7%和38.8%~99.2%;其中与ALV-K参考毒株env基因的核苷酸和氨基酸的相似性较高,分别为93.0%~99.7%和91.9%~99.2%;与ALV-K原型株JS11C1的核苷酸和氨基酸相似性分别为94.9%~97.7%和94.2%~98.5%;与ALV-A、ALV-B、ALV-C、ALV-D、ALV-E、ALV-J参考毒株的核苷酸和氨基酸相似性较低。与ALV-K原型株JS11C1相比,选取的20株ALV-K存在61处突变位点。说明河北省部分地区太行鸡群存在ALV的感染,ALV-J和ALV-K为主要流行的ALV亚群,其env基因变异较大�In order to understand the subpopulation of Avian leukosis virus(ALV)and its genetic and evolutionary characteristics in Taihang chicken flocks in some areas of Hebei Province,in this study,from July 2022 to March 2023,97 mixed samples of liver and spleen tissues of Taihang chickens were collected from 10 farms in Baoding City and Shijiazhuang City,Hebei Province.ALV-A,ALV-B,ALV-J,and ALV-K of these samples were detected by RT-PCR;molecular biology software such as DNASTAR and MegAlign was used to analyze the genetic evolution based on sequencing results of the env gene of some of the detected strains(random selection).The results showed that the total detection rate of ALV in tissue specimen was 60.8%(59/97).Among them,the detection rate of ALV-A and ALV-B was 0;the detection rate of ALV-J was 19.6%(19/97);the detection rate of ALV-K was 55.7%(54/97);the mixed detection rate of ALV-J and ALV-K was 14.4%(14/97).The nucleotide and amino acid similarities of env genes between the 9 ALV-J strains selected were 92.5%-100%and 88.1%-100%,respectively.The nucleotides and amino acids similarities of the env genes between 9 ALV-J strains selected and 15 ALV reference strains were 59.7%-97.6%and 51.0%-97.4%,respectively.The nucleotides and amino acids of the env gene were highly similar to those of the ALV-J reference strain,which were 84.0%-97.6%and 84.9%-97.4%,respectively.The similarity of nucleotides and amino acids with ALV-J prototype strain HPRS103 was 93.0%-95.4%and 88.8%-94.7%,respectively.The nucleotide and amino acid similarities with the ALV-A,ALV-B,ALV-C,ALV-D,ALV-E,and ALV-K reference strains were low.Compared with the ALV-J prototype strain HPRS103,9 ALV-J strains selected had 28 mutation sites and 1 deletion site.The nucleotide and amino acid similarities of env genes between the 20 ALV-K strains selected were 96.0%-99.7%and 94.0%-99.2%,respectively.The nucleotides and amino acids of the env genes of 20 ALV-K strains selected and 11 ALV reference strains were 51.7%-99.7%and 38.8%-99.2%,respectively.Among th
