摘  要:为了探究玉屏风散总多糖佐剂联合鸡新城疫、禽流感二联灭活疫苗胚胎免疫效果,试验将吉林卢花鸡种蛋置于孵化器中孵化,孵化至18.5胚龄时随机分为4组,分别为对照组(Control组)及LⅠ、LⅡ、LⅢ组,每组3个重复,每个重复33枚。Control组和LⅠ组不做任何处理,继续孵化;LⅡ组胚蛋通过羊膜腔注射新城疫、禽流感二联灭活疫苗0.3 mL+生理盐水0.5 mL;LⅢ组胚蛋通过羊膜腔注射注射4 mg/mL玉屏风总多糖0.5 mL+新城疫、禽流感二联灭活疫苗0.3 mL。雏鸡出壳后,LⅠ组雏鸡接种新城疫、禽流感二联灭活疫苗,其他3组正常饲养,分别于雏鸡7,14,21,28日龄时,每个重复随机选取1只雏鸡,采集血液分离血清,检测新城疫和H9亚型禽流感HI抗体效价及细胞因子[白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、白细胞介素-10(IL-10)、γ干扰素(IFN-γ)]质量浓度;分离T淋巴细胞,检测T淋巴细胞中CD3^(+)CD4^(+)、CD3^(+)CD8^(+)T淋巴细胞百分比;在雏鸡14日龄时,每组随机选取70只雏鸡进行新城疫病毒(NDV)和禽流感病毒(AIV)攻毒,攻毒后统计保护率,进行鸡胚半数感染量(EID50)试验,评价免疫保护效果。结果表明:在7,14,21,28日龄时,LⅢ组血清NDV和AIV HI抗体效价显著高于Control组和LⅠ组、LⅡ组(P<0.05)。在7,14,21,28日龄时,LⅢ组血清中IL-2、IL-4、IL-10(除7日龄外)、IFN-γ(除14日龄外)质量浓度均显著高于其他组(P<0.05),且各组血清中IL-4、IL-10、IFN-γ质量浓度均在21日龄时达到最高,28日龄回落,IL-2质量浓度均在28日龄时达到最高。在7,14日龄时,LⅢ组CD3^(+)CD4^(+)、CD3^(+)CD8^(+)T淋巴细胞百分比显著高于其他组(P<0.05);在21,28日龄时,LⅡ组与LⅢ组CD3^(+)CD4^(+)、CD3^(+)CD8^(+)T淋巴细胞百分比差异不显著(P>0.05),但均显著高于LⅠ组和Control组(P<0.05)。LⅠ组和LⅢ组NDV免疫保护率分别为70%和92%,LⅡ组为80%。AIV攻毒后各组雏鸡并未见死亡。LⅢ组NDV和AIV EID50显�In order to expoler the effects of the total polysaccharide adjuvant of Yupingfeng powder combined with inactivated vaccine of Newcastle disease and avian influenza via embryonic immunization,in the test,the eggs of Jilin Barred Plymouth Rocks were incubated in the incubator,and when they were hatched to 18.5 embryonic age,they were randomly divided into 4 groups,which were the Control group and the LⅠ,LⅡ,LⅢgroups,with 3 repetitions in each group and 33 pieces in each repetition.The Control group and LⅠgroup did not do any treatment and continued to hatch;amniotic injection in the LⅡgroup of 0.3 mL of Newcastle disease-avian influenza inactivated vaccine+0.5 mL of normal saline;amniotic injection in the LⅢgroup of 4 mg/mL total polysaccharide of Yupingfeng powder 0.5 mL+Newcastle disease-avian influenza inactivated vaccine 0.3 mL.After the chicks were hatched,the chicks in the LⅠgroup were vaccinated with Newcastle disease-avian influenza inactivated vaccine,and the other three groups were raised normally.At the age of 7,14,21 and 28 days,1 chick was randomly selected from each repetition,blood was collected,and serum was separated.The HI antibody titers of Newcastle disease and H9 subtype avian influenza in chicks,and the mass concentrations of cytokines interleukin(IL)-2,IL-4,IL-10 andγinterferon(IFN-γ)were detected.T lymphocytes were isolated,and the percent of CD3^(+)CD4^(+),CD3^(+)CD8^(+)T lymphocytes in T lymphocytes were detected.At 14 days of age,70 chicks in each group were randomly selected for Newcastle disease virus(NDV)and Avian influenza virus(AIV)challenge.The protection rate was counted after challenge,and the EID50 test was carried out to evaluate the immune protection effect.The results showed that at 7,14,21 and 28 days of age,the serum NDV and AIV HI antibody titers of chicks in the LⅢgroup were significantly higher than those in the Control group and LⅠgroup,LⅡgroup(P<0.05).At 7,14,21 and 28 days of age,the serum mass concentrations of IL-2,IL-4,IL-10(except 7 days old)a
