摘 要:为了探讨不同剂量的复合微生态制剂对断奶仔猪炎性因子与肠道黏膜形态的影响,试验将100头28日龄、体重相近[(10.0±0.5)kg]的断奶仔猪随机分为5组,每组5个重复,每个重复4头。空白对照组(C组)只饲喂基础日粮,Ⅰ、Ⅱ、Ⅲ组饲喂分别添加0.1%、0.2%、0.3%复合微生态制剂的基础日粮,抗生素对照组(A组)饲喂添加75 mg/kg金霉素的基础日粮。预试期7 d,正试期28 d。采用ELISA法检测仔猪血清和肠道(空肠、回肠、结肠)黏膜炎症因子的水平,并采用实时荧光定量PCR法检测肠道黏膜炎症因子的mRNA表达水平;制作十二指肠、空肠和回肠组织石蜡切片并进行H.E.染色,检测仔猪肠道的黏膜形态。结果表明:与C组比较,其余组血清中肿瘤坏死因子-α(TNF-α)质量浓度及A组和Ⅱ、Ⅲ组血清中白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)质量浓度显著降低(P<0.05),且白细胞介素-10(IL-10)质量浓度显著升高(P<0.05);Ⅱ、Ⅲ组空肠、回肠中TNF-α、IL-6和IL-1β含量显著降低(P<0.05),IL-10含量显著升高(P<0.05),结肠中IL-1β含量显著降低(P<0.05);Ⅰ、Ⅱ、Ⅲ组和A组空肠、回肠黏膜TNF-αmRNA相对表达量和Ⅱ、Ⅲ组空肠、回肠、结肠黏膜IL-1βmRNA相对表达量及空肠、回肠黏膜IL-6 mRNA相对表达量均显著降低(P<0.05),Ⅱ组空肠IL-10 mRNA相对表达量显著升高(P<0.05)。与A组比较,Ⅰ组血清中TNF-α质量浓度显著增加(P<0.05),IL-10质量浓度显著降低(P<0.05);Ⅱ、Ⅲ组空肠黏膜中TNF-α含量显著降低(P<0.05);Ⅱ组结肠黏膜IL-1βmRNA相对表达量显著降低(P<0.05),Ⅰ、Ⅱ、Ⅲ组结肠黏膜IL-6 mRNA相对表达量显著增加(P<0.05)。Ⅰ、Ⅱ、Ⅲ组十二指肠的绒毛长度和绒隐比显著高于C组和A组(P<0.05),其中Ⅱ组绒隐比最高;Ⅱ和Ⅲ组隐窝深度显著低于A组(P<0.05)。Ⅱ、Ⅲ组空肠绒毛长度显著高于C组(P<0.05),其中Ⅱ组显著高于A组(P<0.05);Ⅰ、Ⅱ、Ⅲ组隐窝深度显著低于In order to investigate the effects of different doses of compound microecological preparations on inflammatory factors and intestinal mucosal morphology of weaned piglets,one hundred 28-day-old weaned piglets with similar body weight([10.0±0.5]kg)were randomly divided into five groups with five replicates in each group and four piglets in each replicate.The animals in the blank control group(group C)were fed the basal diet.The animals in groupsⅠ,ⅡandⅢwere fed a basal diet supplemented with 0.1%,0.2%and 0.3%compound microecological preparations,respectively.The control group with antibiotic(group A)was supplemented with 75 mg/kg gentamycin in the basal diet.The pre-feeding period was 7 days,and the test period was 28 days.The levels of inflammatory factors in serum and intestinal(jejunum,ileum and colon)mucosa of piglets were detected by ELISA,and the expression levels of inflammatory factors in intestinal mucosa were detected by real-time fluorescence quantitative PCR.The morphology of intestinal mucosa of piglets was observed by paraffin sections of duodenal villi jejunum and ileum tissues and H.E.staining.The results showed that compared with group C,the mass concentration of TNF-αin the serum of other groups,as well as the mass concentration of IL-6 and IL-1βin the serum of group A,groupⅡandⅢwere significantly decreased(P<0.05),and the mass concentration of IL-10 was significantly increased(P<0.05);the contents of TNF-α,IL-6 and IL-1βin the jejunum and ileum in the groupsⅡandⅢwere significantly decreased(P<0.05);the IL-10 content were significantly increased(P<0.05),and IL-1βcontent were significantly decreased in the colon(P<0.05);the relative expression level of TNF-αmRNA in the jejunum and ileum mucosa in the groupsⅠ,Ⅱ,Ⅲand group A,and the relative expression level of IL-1βmRNA in the jejunum,ileum,and colon mucosa and the relative expression level of IL-6 mRNA in the jejunum and ileum mucosa in the groupsⅡandⅢwere all significantly decreased(P<0.05);the relative expression
