摘 要:为确定淮南市潘集区肉牛4种胃肠道线虫的流行和虫种分布情况,从淮南市潘集区多个牛场共采集511份新鲜肉牛粪便样品,采用基于4种胃肠道线虫ITS2基因的半巢式PCR方法对所有样本进行检测,对获得的线虫ITS2基因进行测序和分析,用Mega软件以最大似然法构建基于4种胃肠道线虫ITS2基因的进化树,以进一步确定其虫种。淮南市潘集区肉牛中共检测出13份胃肠道线虫阳性样本,总感染率为2.5%(13/511),共检出1份古柏属、6份血矛属、4份食道口属和2份毛圆属线虫,感染率分别为0.2%(1/511)、1.2%(6/511)、0.8%(4/511)和0.4%(2/511),未检出混合感染样本。序列及进化树分析显示本次获得的胃肠道线虫虫种分别为捻转血矛线虫、点状古柏线虫、哥伦比亚食道口线虫和蛇形毛圆线虫。潘集区肉牛胃肠道线虫感染率较低,显示肉牛养殖中驱虫工作做得比较好,但考虑到胃肠道线虫的危害及人兽共患虫种的存在,仍然要高度重视其防控工作。To determine the prevalence and distribution of four gastrointestinal nematodes in beef cattle in Panji District,Huainan City.A total of 511 fresh fecal samples were collected from multiple cattle farms in Panji District,Huainan City.The samples were tested using a semi-nested PCR method based on the ITS2 gene of four gastrointestinal nematodes.The ITS2 genes obtained from the four gastrointestinal nematodes were sequenced and analyzed,and the evolutionary tree based on the ITS 2 gene of the four gastrointestinal nematodes was constructed using Mega software with the maximum likelihood method to further verify the species of the four gastrointestinal nematodes obtained in this study.A total of 13 positive samples of gastrointestinal nematodes were detected in beef cattle in Panji District,Huainan City.The total infection rate of gastrointestinal nematodes was 2.5%(13/511).One Cooperia spp.,six Haemonchus spp.,four Oesophagostomum spp.,and two Trichostrongylus spp.were identified by the semi-nested PCR,with infection rates of 0.2%(1/511),1.2%(6/511),0.8%(4/511),and 0.4%(2/511),respectively.No mixed infection were detected.Sequence and phylogenetic analysis revealed that the gastrointestinal nematode species obtained in this study were Haemonchus contortus,Cooperia punctata,Oesophagostomum columbianum,and Trichostrongylus colubriformis.The infection rate of gastrointestinal nematodes in beef cattle in Panji District is relatively low,indicating that the deworming work in beef cattle breeding is done well.However,considering the harm of gastrointestinal nematodes and the presence of zoonotic species,it is still necessary to attach great importance to their prevention and control work.
