摘 要:为建立鸡胚的小肠上皮细胞(Intestinal Epithelial Cells,IECs)体外培养方法,采用0.25%胰蛋白酶-EDTA消化法,分离鸡胚的IECs,然后通过细胞筛、相差贴壁法纯化分离到的鸡IECs,最后用碱性磷酸酶染色法对分离的细胞进行鉴定。通过胰蛋白酶消化法,分离得到鸡IECs,经碱性磷酸酶染色后细胞均为黑色,表明细胞筛、相差贴壁法纯化法对鸡IECs纯化效果较好,纯化后的细胞均为鸡IECs。体外培养结果表明,分离到的鸡IECs能够在体外条件下增殖培养8 d。本试验从鸡胚小肠组织中成功分离纯化到了IECs,且能在体外条件下进行增殖培养。The intestinal epithelial cells(IECs)of chicken embryos were isolated in vitro,which provided a reference for further research on the pathogenesis of chicken intestinal diseases.The isolation of chicken embryo IECs was achieved using the 0.25%trypsin-EDTA digestion method,followed by purification through cell screening and phase difference adhesion method.Finally,the isolated cells were identified by alkaline phosphatase staining method.Chicken IECs were isolated by trypsin digestion,and all the cells were black-stained by alkaline phosphatase,which indicated that the purified cells were chicken IECs.Moreover,these isolated chicken IECs demonstrated an ability to proliferate in vitro for up to 8 days.In this experiment,IECs were successfully isolated from chicken small intestine tissue,and they could be cultured for proliferation in vitro.
