摘 要:目的探究四物汤(Siwu tang,SWT)对自然衰老小鼠皮肤和机体的改善作用及其可能的机制。方法将自然衰老小鼠随机分为模型组、阳性注射对照组、阳性外用对照组、SWT内服给药组、SWT外用组、SWT内外合用给药组,另设三月龄空白对照组,连续给药24天。检测小鼠血浆超氧化物歧化酶(SOD)活性、过氧化物酶(POD)活性和过氧化氢酶(CAT)活性以及皮肤组织超氧化物歧化酶(SOD)活性和过氧化氢酶(CAT)活性,通过HE染色观察小鼠用药皮肤组织病理形态学变化。结果与模型组比较,SWT外用组小鼠皮肤组织中SOD和CAT活性显著升高(P<0.05);SWT内服给药组小鼠血浆SOD、CAT和POD活性显著升高(P<0.05);皮肤组织病理形态学显示SWT外用组和SWT内外合用给药组小鼠皮肤毛囊皮脂腺数量明显增加,表皮层真皮层均变厚且分界重新清晰。结论SWT能通过提高皮肤和机体的抗氧化能力、增强皮肤弹性,从而改善自然衰老小鼠的皮肤状态,有效的延缓和改善机体衰老过程。Objective The aim is to explore the potential of Siwu Tang(SWT)in enhancing the skin and body of mice in their natural aging stages.Method Naturally aging mice were randomly assigned to a model group,a positive injection control group,a positive external control group,a SWT oral administration group,a SWT emulsion external administration group,a SWT combined internal and external administration group,and a three-month-old blank control group.The treatment was continuously carried out for 24 days.The activities of mouse plasma superoxide dismutase(SOD),peroxidase(POD),and catalase(CAT),as well as the activities of skin tissue superoxide dismutase(SOD)and catalase(CAT)were detected.Additionally,the pathological and morphological changes of the drug-treated skin tissue of mice were observed through HE staining.Results Showed that compared with the model group,the activities of SOD and CAT in the skin tissue of mice in the SWT emulsion external administration group were significantly increased(P<0.05).The activities of plasma SOD,CAT,and POD in mice in the SWT oral administration group were significantly increased(P<0.05).In terms of pathological morphology of skin tissue,it was observed that the number of pilosebaceous glands in the skin of mice in the SWT emulsion external administration group and the SWT combined internal and external administration group increased significantly,and the epidermis and dermis became thicker and the boundaries became clearer once again.Conclusion SWT can effectively delay and improve aging in mice by enhancing the antioxidant capacity of the skin and body and improving skin elasticity.
