摘 要:目的:从中药白及中提取白及多糖(BSP),并用硫辛酸(LA)进行疏水改性,得到两亲性聚合物胶束,对其制备工艺进行优化和评价,并以抗肿瘤药物姜黄素(Cur)为模型药物,探究其作为药物载体的潜力。方法:超声提取法提取BSP,通过活性炭脱色素、sevag法除蛋白纯化。利用粒径、PDI为评价指标,通过单因素试验优化制备工艺,得到胶束的最佳制备方案;以载药量和包封率为评价指标,对其载药方法进行筛选和优化。结果:BSP-LA聚合物胶束既具有亲水性也具有疏水性,平均粒径为(172.4±1.60)nm,平均PDI为0.207±0.01,可作为一种良好的药物载体,Cur载药量为4.13%,包封率为41.3%。结论:通过LA对BSP进行疏水改性可制备出粒径适宜的聚合物胶束,并具有较高的药物装载率,拓宽了可用于BSP改性材料的范围,同时为其进一步作为药物载体用于抗肿瘤研究提供了实验基础。Objective:To obtain amphiphilic polymer micelles by extracting Bletilla striata polysaccharide from Bletilla striata and using lipoic acid for hydrophobic modification.The preparation process was optimized and evaluated.Curcumin,an antitumor drug,was used as a model drug to explore its potential as drug carrier.Methods:BSP were extracted by ultrasonic extraction and purified by activated carbon depigmentation and sevag deproteinization.Using particle size and PDI as evaluation indexes,the preparation process was optimized by single factor test to obtain the best preparation scheme of micelles.The drug loading and encapsulation rate were used as evaluation indexes to screen and optimize the drug loading methods.Results:The average particle size was(172.4±1.60)nm,and the average PDI was 0.207±0.01.It could be used as a good drug carrier.The drug carrying capacity of curcumin was 4.13%,and the encapsulation rate was 41.3%.Conclusion:Polymer micelles with suitable particle size can be prepared by hydrophobic modification of BSP with high drug loading rate,which broadens the range of materials that can be used for modification of BSP and provides experimental basis for their further use as drug carriers in anti-tumor research.
