不同生物型牛病毒性腹泻病毒NS4B蛋白的生物信息学分析

摘  要:牛病毒性腹泻病毒(BVDV)是一种引起牛病毒性腹泻的高度致死性病原,根据生物型不同可分为致细胞病变型(CP)及非致细胞病变型(NCP)2种。本研究通过多种生物信息学方法,对这2种生物型BVDV毒株NS4B蛋白的差异及相似性进行了比较。结果显示:2种生物型BVDV的NS4B蛋白均属于不含信号肽和跨膜区的不稳定性疏水蛋白,其N端具有较高变异性;亚细胞定位预测发现,NS4B蛋白不含有线粒体导肽(mTP)或分泌通道信号肽(SP),且定位在内质网和线粒体中的概率最高;高级结构预测发现,2种生物型NS4B蛋白的二级与三级结构相似,均以α-螺旋为主;B细胞抗原表位分析发现,NCP型BVDV含有8个B细胞抗原表位肽段,CP型含有9个;蛋白翻译后修饰预测发现,NCP型BVDV具有1个特异性N-糖基化修饰位点,NCP型BVDV具有36个潜在的磷酸化修饰位点,而CP型具有35个。结果表明:2种生物型BVDV NS4B蛋白的基本理化性质及蛋白质结构相似性较高,但亚细胞定位情况和蛋白翻译后修饰有所不同,推测N端氨基酸序列的高突变性及蛋白翻译后修饰的不同可能会影响不同生物型BVDV与发病动物临床症状之间的关系。Bovine viral diarrhoea virus(BVDV)is a highly lethal pathogen causing bovine viral diarrhoea(BVD),and could be classified into cytopathic(CP)and non-cytopathic(NCP)types based on different biotypes.In the study,differences and similarities between NS4B proteins of the two biotypes were compared by various bioinformatics methods.The results showed that both the NS4B proteins belonged to unstable hydrophobic proteins without signal peptides and transmembrane regions,and high variability was observed on their N terminal;subcellular localization prediction showed that mitochondrial conduction peptide(mTP)or secretory channel signaling peptide(SP)was not contained in NS4B protein that was located in endoplasmic reticulum and mitochondria with a highest probability;advanced structure prediction showed that the secondary and tertiary structures of NS4B proteins of the two biotypes were similar,and dominated byα-helix;B-cell epitope analysis showed that 8 B-cell epitope peptides were contained in BVDV NCP biotype,while 9 were contained in CP biotype;protein post-translational modification prediction showed that 1 specific N-glycosylation modification site and 36 potential phosphorylation modification sites were observed in BVDV NCP biotype,while 35 phosphorylation modification sites were in CP biotype.In conclusion,the general physicochemical properties and protein structures of NS4B proteins of the two biotypes were highly similar,but the subcellular localization and protein post-translational modification were different,speculating that the relationship between BVDV with different biotypes and clinical symptoms of infected animals could be affected by high mutation of N-terminal amino acid sequence and the difference of protein post-translational modification.
