摘 要:目的研究并建立金嗓颗粒的薄层鉴别与黄酮类成分含量测定分析方法,为该制剂质量标准的建立奠定基础。方法薄层色谱法(TLC)对金嗓颗粒中黄芩药材进行定性鉴别,紫外-可见分光光度法(UV-Vis)测定金嗓颗粒中总黄酮的含量,高效液相色谱法(HPLC)测定金嗓颗粒中黄芩苷、黄芩素、汉黄芩苷和汉黄芩素含量。结果UV-Vis显示,金嗓颗粒中总黄酮类成分的最大吸收波长为276 nm,平均回收率(n=6)为100.11%。TLC显示,金嗓颗粒与对照品在相应位置斑点清晰、分离度好,阴性对照无干扰。HPLC显示,黄芩苷、黄芩素、汉黄芩苷和汉黄芩素线性关系均良好,r≥0.9995;平均回收率(n=6)分别为96.87%、93.10%、100.65%与99.37%,均符合规定的回收率限度。结论本实验建立的金嗓颗粒中黄芩药材定性鉴别和黄酮类成分定量分析方法准确可靠、专属性强、重现性好,为金嗓颗粒的质量评价提供科学依据。Objective To study and establish the analytical method of thin-layer identification and determination of flavonoid content of Jinsang Granules,and to lay the foundation for the establishment of the quality standard of this preparation.Methods Thin-layer chromatography(TLC)was used for the qualitative identification of Scutellaria baicalensis in Jinsang Granules,ultraviolet-visible spectrophotometry(UV-Vis)for the determination of total flavonoids,and high performance liquid chromatography(HPLC)for the determination of baicalin,baicalein,wogonoside,wogonin in Jinsang Granules.Results UV-Vis showed that the maximum absorption wavelength of the total flavonoids in Jinsang Granules was 276 nm,and the average recovery(n=6)were 100.11%.TLC showed that the spots of Jinsang Granules and the control at the corresponding positions were clear and well separated,and the negative control was not interfered with.HPLC showed that the linearity of baicalin,baicalein,wogonoside,wogonin were good,with r≥0.9995,the average recoveries(n=6)were 96.87%,93.10%,100.65% and 99.37%,respectively,which met the specified recovery limits.The average recoveries(n=6)were 96.87%,93.10%,100.65% and 99.37%,respectively,which were in accordance with the specified recovery limits.Conclusion The qualitative identification of Scutellaria baicalensis and quantitative analysis of flavonoids in Jinsang Granules established in this experiment are accurate,reliable,exclusive and reproducible,which provide scientific basis for the quality evaluation of Jinsang Granules.
