摘 要:【目的】观察下气汤对结肠癌化疗相关性癌因性疲乏小鼠的治疗作用及机制。【方法】将成功构建的结肠癌化疗相关性癌因性疲乏模型小鼠随机分为模型对照组和下气汤组,每组10只,另设空白对照组。每组给予相应干预后,测定小鼠力竭游泳时间,测量肿瘤体积,计算抑瘤率,采用酶联免疫吸附分析(ELISA)检测血清中肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β、IL-6等炎症因子及丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、尿素氮(BUN)等安全性指标水平,采用Western Blot法检测瘤块(或腋下组织)上皮细胞中Toll样受体4(TLR4)、核转录因子κB(NF-κB)p65蛋白表达水平。【结果】(1)治疗前,模型对照组小鼠力竭游泳时间显著短于空白对照组(P<0.01);治疗后第28天,下气汤组小鼠力竭游泳时间显著长于模型对照组(P<0.01),且具有时间依赖性。(2)下气汤组小鼠治疗4~28 d瘤体体积逐渐减小,治疗28 d后的抑瘤率为68.96%。(3)模型对照组小鼠血清中TNF-α、IL-1β、IL-6水平显著高于空白对照组(P<0.01);下气汤组血清中TNF-α、IL-1β、IL-6水平显著低于模型对照组(P<0.05)。(4)模型对照组小鼠血清中ALT、AST、BUN水平均高于空白对照组(P<0.05或P<0.01);下气汤组血清中ALT、AST、BUN水平均低于模型对照组(P<0.05)。(5)模型对照组瘤块上皮细胞TLR4、NF-κB p65蛋白表达水平均高于空白对照组(P<0.01);下气汤组瘤块上皮细胞TLR4、NF-κB p65蛋白表达水平低于模型对照组(P<0.01)。【结论】下气汤可有效促进结肠癌化疗相关性癌因性疲乏小鼠的体能恢复,抑制肿瘤增长,其机制可能与抑制TLR4、NF-κB p65表达,进而减轻炎症反应有关,且该方具有一定的安全性。Objective To observe the therapeutic effect and mechanism of Xiaqi Decoction for mice with chemotherapy-induced cancer-related fatigue(CRF)in colon cancer.Methods The successfully constructed chemotherapy-induced CRF mouse model of colon cancer was randomly divided into the model control group and the Xiaqi Decoction group,with 10 mice in each group,and a blank control group was also set up.After each group was given the corresponding intervention,the mice exhaustive swimming time was determined,the tumor size was measured,the tumor inhibition rate was calculated,and the serum levels of inflammatory factors such as tumor necrosis factor(TNF)-α,interleukin(IL)-1β,IL-6 and other safety indicators such as alanine aminotransferase(ALT),aspartate aminotransferase(AST)and blood ureanitrogen(BUN)were detected by enzyme-linked immunosorbent assay(ELISA),the protein expression levels of Toll-like receptor 4(TLR4)and nuclear factorκB(NF-κB)p65 in the epithelial cells of tumor lumps(or axillary tissues)were detected by Western Blot.Results(1)Before treatment,the exhaustive swimming time of mice in the model control group was significantly less than that of the blank control group(P<0.01);after 28 days of treatment,the exhaustive swimming time of mice in the Xiaqi Decoction group was significantly longer than that of the model control group(P<0.01),and it was time-dependent.(2)The tumor size of mice in the Xiaqi Decoction group was gradually decreased from 4-28 days of treatment,and the tumor inhibition rate was 68.96%after 28 days of treatment.(3)The serum levels of TNF-α,IL-1βand IL-6 in the model control group were significantly higher than those in the blank control group(P<0.01);and the serum levels of TNF-α,IL-1βand IL-6 in the Xiaqi Decoction group were significantly lower than those in the model control group(P<0.05).(4)The serum levels of ALT,AST and BUN in the model control group were higher than those in the blank control group(P<0.05 or P<0.01);the serum levels of ALT,AST and BUN in the Xiaqi Decoction gr
