山桐子油抗氧化活性及其对HepG2非酒精性脂肪性肝病脂质蓄积的影响研究

摘  要:为提高山桐子(Idesia polycarpa Maxim.)油的应用价值,在探究山桐子油抗氧化活性的基础上,采用油酸(oleic acid, OA)诱导人肝癌(human hepatocellular carcinoma, HepG2)细胞建立体外高脂类非酒精性脂肪性肝病(nonalcoholic fatty liver disease, NAFLD)模型,并分析山桐子油对HepG2细胞NAFLD模型脂质蓄积的影响。结果表明,冷榨山桐子油表现出较强的抗氧化能力,并对NAFLD模型HepG2细胞内脂滴的大小、数量、细胞的损伤程度都产生明显影响。处理后的HepG2细胞甘油三酯(triglyceride, TG)和总胆固醇(total cholesterol, TC)含量、谷草转氨酶(aspartate transaminase, AST)和谷丙转氨酶(alanine transaminase, ALT)活性明显下降。山桐子油通过增强抗氧化活性、减少肝损伤、调节脂质代谢对OA诱导HepG2细胞肝脂肪变性具有明显的缓解作用。该研究结果可为山桐子油作为功能油脂的开发利用提供科学依据。In order to improve application value of Idesia polycarpa Maxim.oil,based on investigating the antioxidant activity of Idesia polycarpa Maxim.oil,oleic acid(OA)was used to induce human hepatocellular carcinoma(HepG2)cells to establish an in vitro high-fat nonalcoholic fatty liver disease(NAFLD)model,and effects of Idesia polycarpa Maxim.oils on lipid accumulation in the NAFLD model of HepG2 cells were carried out.The results showed that cold-pressed Idesia polycarpa Maxim.oil showed its strong antioxidant capacity,and had a significant effect on the size and number of lipid droplets in NAFLD model of HepG2 cells and the degree of cellular damage.The contents of triglyceride(TG)and total cholesterol(TC),the activities of aspartate transaminase(AST)and alanine transaminase(ALT)in the treated HepG2 cells were significantly decreased.Idesia polycarpa Maxim.oil had a significant mitigating effect on OA-induced fatty degeneration of liver in HepG2 cells by enhancing antioxidant activity,reducing liver injury,and regulating lipid metabolism.The results provided a scientific basis for the development and utilization of as a functional lipid.
