摘 要:目的观察半夏对慢性阻塞性肺疾病(COPD)大鼠EGFR/ERK/Sp1/MUC5AC通路的影响,探讨半夏治疗COPD可能的分子机制。方法SD大鼠60只,10只为空白组,50只建立COPD大鼠模型,随机分为模型组、阳性对照组及不同剂量半夏治疗组(半夏L组、半夏M组、半夏H组)。空白组和模型组给予生理盐水灌胃,阳性对照组给予地塞米松溶于生理盐水灌胃,半夏L组、半夏M组、半夏H组分别给予半夏提取物10、30、60 g/kg(相当于生药剂量)灌胃,干预4周后,比较各组大鼠肺灌洗液和肺组织中肿瘤坏死因子-α(TNF-α)、白介素(IL)-1β、IL-6含量,肺组织病理切片及EGFR、ERK、Sp1、MUC5AC蛋白表达量。结果半夏可显著改善COPD大鼠受损的肺功能,降低COPD大鼠肺灌洗液和肺组织中TNF-α、IL-1β、IL-6含量,且均呈现良好的量效关系;Western blot法、免疫组织化学法和免疫荧光法检测结果表明,半夏可显著降低COPD大鼠肺组织中EGFR、ERK、Sp1、MUC5AC蛋白表达量(P<0.05或P<0.01)。结论半夏可显著改善COPD大鼠的肺功能,其机制是抑制高度活化的EGFR/ERK/Sp1/MUC5AC通路。Objective To observe the effects of pinellia on EGFR/ERK/Sp1/MUC5AC pathway in chronic obstructive pulmonary disease(COPD)rats,and to explore the possible molecular mechanism of pinellia in treating COPD.Methods A total of 60 SD rats,10 of which were blank group and 50 COPD rats,were randomly divided into model group,positive control group and pinellia L group,pinellia M group and pinellia H group.Blank group and model group were given normal saline intragastric administration,positive control group was given dexamethasone dissolved in normal saline intragastric administration,pinellia L group,pinellia M group and pinellia H group were given pinellia extract 10,30,60 g/kg(equivalent to crude drug dose)intragastric administration,4 weeks after intervention,the contents of TNF-α,IL-1βand IL-6 in lung lavage fluid and lung tissue,pathological sections of lung tissue and expression levels of EGFR,ERK,Sp1 and MUC5AC were compared.Results Pinellia could significantly improve the impaired lung function of COPD rats,and reduced the contents of TNF-α,IL-1βand IL-6 in lung lavage fluid and lung tissue of COPD rats,and all showed a good dose-effect relationship.The results of Western blot,immunohistochemical method and immumofluorescence method showed that pinellia could significantly reduce the protein expression levels of EGFR,ERK,Sp1 and MUC5AC in lung tissue of COPD rats(P<0.05 or P<0.01).Conclusion Pinellia can significantly improve lung function in COPD rats by inhibiting the highly activated EGFR/ERK/Sp1/MUC5AC pathway.
