摘  要:以从辽宁环渤海湾土壤中筛选获得的1株放线菌为发酵菌株,以发酵产物米卡芬净母环前体含量为评价指标,通过Plackett-Burman实验筛选出麸质粉、KH_(2)PO_(4)和MgSO_(4)等3个影响发酵的主要因素;通过最陡爬坡实验确定Box-Behnken响应面分析的水平中心点,通过响应面分析得到最佳培养基组分(g·L^(-1))为:葡萄糖10、山梨醇160、麸质粉20.05、棉籽饼粉10、(NH_(4))_(2)SO_(4)12、KH_(2)PO_(4)5.25、MgSO_(4)0.92、MnSO_(4)0.5、CuSO_(4)1、CaCO_(3)8,pH值6.5。在此条件下,发酵液中米卡芬净母环前体含量达到5956.37 mg·L^(-1),比优化前提高了1.12倍。Taking a actinomycete screened from the soil around Bohai Bay in Liaoning province as a fermentation strain,and the content of Micafungin mother ring precursor as an evaluation index,we screened out three main factors affecting fermentation,including gluten powder,KH2PO4,and MgSO4 by Plackett-Burman experiments.Moreover,we determined the horizontal center point of Box-Behnken response surface analysis through the steepest ascent experiments,and determined the optimal medium component(g·L^(-1))through response surface analysis as follows:the glucose of 10,the sorbitol of 160,the gluten powder of 20.05,the cottonseed cake powder of 10,the(NH_(4))_(2)SO_(4) of 12,the KH_(2)PO_(4) of 5.25,the MgSO_(4) of 0.92,the MnSO_(4) of 0.5,the CuSO_(4) of 1,the CaCO_(3) of 8,and the pH value of 6.5.Under above conditions,the content of Micafungin mother ring precursor in fermentation solution reaches 5956.37 mg·L^(-1),which is 1.12 times higher than that before optimization.
