摘  要:选用5种大孔树脂对板栗外皮和内皮结合态多酚(BP)进行纯化,以吸附率和解吸率为评价指标,对大孔树脂进行了筛选,并对纯化工艺进行了优化。结果表明,采用NKA-9大孔树脂,板栗外皮BP(COSBP)的最佳纯化条件如下:上样浓度为0.22 mg·mL^(-1)、上样pH值为3、上样流速为4.0 BV·h^(-1)、上样体积为26 mL,洗脱液为70%乙醇、洗脱液pH值为7、洗脱流速为3.0 BV·h^(-1)、洗脱体积为28 mL;板栗内皮BP(CISBP)的最佳纯化条件如下:上样浓度为1.33 mg·mL^(-1)、上样pH值为3、上样流速为0.5 BV·h^(-1)、上样体积为8 mL,洗脱液为70%乙醇、洗脱液pH值为7、洗脱流速为0.5 BV·h^(-1)、洗脱体积为33 mL;在上述纯化条件下,COSBP纯度从(1.40±0.04)%升至(24.10±0.35)%,CISBP纯度从(5.00±0.03)%升至(30.50±0.14)%。该工艺绿色环保,为板栗皮BP的进一步利用提供了参考依据。We selected five kinds of macroporous resins to purify the bound polyphenols(BP)from outer shell and inner shell of chestnut.Taking adsorption rate and desorption rate as evaluation indexes,we screened the macroporous resins,and optimized the purification process.The results show that NKA-9 macroporous res-ins are adopted,and the optimal purification conditions of chestnut outer shell BP(COSBP)are as follows:the sample concentration is 0.22 mg·mL^(-1),the sample pH value is 3,the sample flow rate is 4.0 BV·h^(-1),the sample volume is 26 mL;70%ethanol is adopted as the eluent,the eluent pH value is 7,the eluent flow rate is 3.0 BV·h^(-1),and the eluent volume is 28 mL.The optimal purification conditions of chestnut inner shell BP(CISBP)are as follows:the sample concentration is 1.33 mg·mL^(-1),the sample pH value is 3,the sample flow rate is 0.5 BV·h^(-1),the sample volume is 8 mL;70%ethanol is adopted as the eluent,the eluent pH value is 7,the eluent flow rate is 0.5 BV·h^(-1),and the eluent volume is 33 mL.Under above purification conditions,the purity of COSBP increases from(1.40±0.04)%to(24.10±0.35)%,and the purity of CISBP increases from(5.00±0.03)%to(30.50±0.14)%.The process is green and environmentally friendly,which provides a ref-erence for the further utilization of chestnut shell BP.
