摘  要:Sox基因是在个体发育过程中具有多种调控功能,编码蛋白中均含HMG(High Mobility Group)基序保守区的基因家族.花(鱼骨)(Hemibarbus maculatus)是在我国广泛分布的一种具有很大养殖潜力的鲤科鱼类.本研究通过同源比对法,从花(鱼骨)转录组数据中鉴定出23个Sox基因序列,分属于B、C、D、E、F 5个亚族.以Sox2、Sox3和Sox4基因为分子钟标记,推测出花(鱼骨)和斑马鱼(Danio rerio)的分化时间约为58.75百万年前.通过RT-PCR对Sox基因在雌雄性腺中表达分析的结果显示,Sox4a、Sox4b、Sox7、Sox8b、Sox9b、Sox13、Sox19b、Sox21a、Sox21b基因在卵巢中特异表达,而Sox1b和Sox12在精巢中特异表达.研究结果将为花(鱼骨)Sox家族的深入研究提供基础资料,同时为进一步探究Sox基因在性别决定与性腺发育中的作用提供重要参考.SRY-related HMG-box(Sox)genes are a family of genes that have multiple regulatory functions in the process of henogenesis,with their encoded proteins containing conserved high mobility group(HMG)motifs.Hemibarbus maculates is a potential aquaculture fish that widely distributes in China.In this study,a total of 23 Sox genes that belong to five subgroups,including B,C,D,E and F,were identified from transcriptome data.The divergent time between Hemibarbus maculates and zebrafish was deduced to be 58.75 million years ago,using Sox2,Sox3 and Sox4 as molecular clock markers.The results of RT-PCR showed that Sox4a,Sox4b,Sox7,Sox8b,Sox9b,Sox13,Sox19b,Sox21a and Sox21b were specifically expressed in ovary,while Sox1b and Sox12 were specifically expressed in testis.The results obtained in this study will provide basic information for further studies on Sox genes,and will be helpful for studies on functions of Sox genes during sex determination and differentiation in this fish.
