益智果实挥发油对慢性束缚应激抑郁小鼠影响的研究

摘　　要：为探讨益智果实挥发油对慢性束缚应激抑郁小鼠的影响,将60只雄性昆明小鼠随机分为空白对照组(不做处理)、慢性束缚应激抑郁模型组(CRS组)、氟西汀组和益智果实挥发油高、中、低剂量组(1.00,0.75,0.50mL/kg)共6组(每组10只).研究显示:与模型组相比,益智果实挥发油高、中、低剂量组强迫游泳不动时间显著缩短,空白对照组旷场实验进入中心区的时间以及益智果实挥发油高、低剂量组和氟西汀组高架十字迷宫实验开臂区的滞留时间均显著延长,益智果实挥发油高、中、低剂量组与氟西汀组SOD活性、GFAP阳性率显著增加,神经细胞形态学相对正常.研究表明,益智果实挥发油可以降低慢性束缚应激抑郁小鼠的抑郁水平,其作用机制可能与益智果实挥发油通过提高SOD活性,从而可减少小鼠体内自由基的堆积和减轻氧化损伤以及增加海马神经元的再生有关.In order to investigate the effect of Alpinia oxyphylla volatile oil on chronic restraint stress depressed mice,60 male Kunming mice were randomly divided into 6 groups of Alpinia oxyphylla volatile oil high,medium and low dose group(1.00,0.75,0.50 mL/kg),chronic restraint stress depression model group(CRS group),blank control group(no treatment)and fluoxetine group in the study,1o mice in each group.The study showed that compared with the model group,the forced swimming immobility time was significantly shortened in the high,medium and low dose groups of Alpinia oryphylla volatile oil,the time to enter the central area of the open field experiment in the blank control group,and the retention time in the open-arm area of the elevated cross maze experiment in the high,low dose groups of Alpinia oryphylla volatile oil and fluoxetine group were all significantly prolonged,the SOD activity and the GFAP positivity rate of the Alpinia oryphylla volatile oil and fluoxetine group by gavage were significantly increased,and the neuronal cell.The morphology of nerve cells was relatively normal.This suggests that the volatile oil of Alpinia oryphylla can reduce the level of depression in mice with chronic bound stress depression,and its mechanism of action may be related to the fact that the volatile oil of Alpinia oxyphylla fruit enhances SOD activity and thus reduce the accumulation of free radicals in mice,reduce oxidative damage and increase the regeneration of hippocampal neurons,thus reducing the depressive symptoms.
