摘  要:目的旨在探讨孕期暴露于炭黑纳米颗粒(carbon black nanoparticles,CBNPs)对F1代小鼠神经系统的潜在影响,尤其是对F1代小鼠的神经损伤及其可能的机制。方法将健康昆明(kunming,KM)种雌性小鼠按体重随机分为对照组(生理盐水组)、低剂量组(2.5 mg/kg)、中剂量组(10 mg/kg)和高剂量组(40 mg/kg)。从妊娠第2天(gestational day 2,GD2)起,对孕鼠进行气管滴注染毒(50μL/只),每隔一天一次,直至GD20。在F1代小鼠出生后第21天(birth day 21,BD21),进行Morris水迷宫实验、脏器指数计算及HE染色观察肺组织和海马区病理变化,同时检测外周血线粒体DNA(mtDNA)拷贝数和血浆超氧化物歧化酶(superoxide dismutase,SOD)活性。结果与对照组相比,低剂量组和高剂量组F1代小鼠在BD2和BD20的体重显著增加,差异具有统计学意义(F=2.761,P<0.05)。高剂量组F1代小鼠的肺(F=5.237,P=0.002)和脑(F=5.033,P=0.003)脏器指数显著高于对照组。HE染色显示高剂量组肺组织结构紊乱、炎症细胞浸润显著,以及海马区锥体细胞排列松散、形态异常。Morris水迷宫实验结果表明,高剂量组小鼠的平均游泳速度显著增加(F=5.113,P=0.003),但平台潜伏期及游泳总路程变化无统计学意义。此外,CBNPs的宫内暴露引起F1代小鼠血浆SOD活性显著降低,外周血mtDNA拷贝数呈下降趋势,但组间差异无统计学意义(F=0.868,P=0.579)。结论孕期CBNPs暴露可能通过引起认知功能受损和抗氧化能力降低对下一代健康构成长期风险,提示其潜在的环境危害需要引起高度关注。Objective The purpose was to investigate effects of prenatal exposure to carbon black nanoparticles(carbon black nanoparticles,CBNPs)on the nervous system of F1 generation mice,and its possible mechanisms.Methods Healthy female Kunming(KM)mice were randomly divided into four groups:control(saline),low-dose(2.5 mg/kg),medium-dose(10mg/kg),and high-dose(40 mg/kg).From day 2 of gestation(GD2),pregnant mice were given tracheal instillation of PrCBNPs(50μL/mouse)every other day until day 20 of gestation(GD20).On postnatal day 21(PND21),F1 offspring underwent Morris water maze tests,organ index calculations,and histopathological evaluation of lung and hippocampal tissues using hematoxylin-eosin(HE)staining.Peripheral blood mitochondrial DNA(mtDNA)copy number and plasma superoxide dismutase(SOD)activity were also measured.Results In contrast to the control group,the body weights of F 1generation mice in the low-dose and high-dose groups exhibited a significant increase on the 2nd and 20th days of birth(F=2.761,P<0.05).Their organ indices of the lungs(F=5.237,P=0.002)and brains(F=5.033,P=0.003)in the high-dose group were significantly higher than those of the control group.HE staining revealed that the lung tissue structure in the high-dose group was disorganized,with marked inflammatory cell infiltration,and the pyramidal cells in the hippocampal region were loosely arranged and morphologically abnormal.The results of the Morris water maze experiment demonstrated that the average swimming speed of mice in the high-dose group significantly increased(F=5.113,P=0.003),but there were no significant changes in the platform latency and total swimming distance.These F1 mice also showed a significant reduction in plasma SOD activity and a downward trend but non-significant changes in the copy number of mtDNA in peripheral blood(F=0.868,P=0.579).Conclusion CBNPs exposure during pregnancy posed health risks to the next generation through impaired cognitive function and reduced antioxidant capacity,suggesting that their potential
