摘　　要：采用海藻酸钠和聚乙烯醇混合物为载体,对产右旋糖酐蔗糖酶的重组大肠杆菌BL21(DE3)/pET-22b-dsrV的固定化条件及其固定化细胞转化蔗糖制备低分子量右旋糖酐的工艺条件进行了研究。实验结果表明:当固定化载体中的海藻酸钠含量为4%,聚乙烯醇含量为1%,菌体包埋量为0.1 g(湿重)/mL时,固定化细胞的效果最好。通过单因素和正交实验表明:固定化细胞最佳转化条件为蔗糖浓度30%、温度为30℃和pH为5.8,固定化细胞具有良好的操作稳定性,可连续使用9批次,蔗糖转换率保持在90%以上,右旋糖酐含量在42%以上。The study investigated the immobilization conditions of recombinant Escherichia coli BL21(DE3)/pET-22b-dsrV producing dextran sucrase using a mixture of sodium alginate(SA)and polyvinyl alcohol(PVA)as carriers.Additionally,the process conditions for the transformation of sucrose into low molecular weight dextran by the immobilized cells were studied.The results showed that the best immobilization effect was achieved when the SA,PVA and embedded cell amount in the immobilization carrier were 4%,1%,and 0.1 g cells(wet weight)/mL,respectively.Through single-factor and orthogonal experiments,it is concluded that the optimal transformation conditions for immobilized cells are sucrose concentration of 30%,temperature of 30°C and pH of 5.8.The immobilized cells possessed excellent operation stability and can continuously convert 9 batches of sucrose,the sucrose conversion rate remains above 90%,and the dextran content is above 42%.
