摘  要:miRNA(microRNA)是一类由为21~23个核苷酸组成的小型非编码RNA分子,它们主要通过靶mRNA序列中3′非编码区(3′UTR)相互结合,进而调控靶基因的表达,一般通过抑制的方式影响靶基因的翻译。目前,miRNA已被证实在机体的各种生命活动过程中发挥了关键的调控作用。因此,研发具有高特异性和高灵敏度的miRNA检测技术尤为重要。该文将围绕Northern blot(Northern印记)、实时荧光定量PCR(qRT-PCR)、荧光原位杂交(FISH)、miRNA测序法进行综述。miRNAs(microRNAs)are small non-coding RNA molecules approximately 21 to 23 nucleotides in length.They primarily regulate target gene expression by binding to the 3′untranslated region(3′UTR)of target mRNA sequences,often suppressing the translation process.Extensive research has demonstrated that miRNAs play crucial regulatory roles in various biological activities.Consequently,developing miRNA detection technologies with high specificity and sensitivity has become a critical focus.This review provides a comprehensive overview of current miRNA detection methods,including Northern blot,quantitative real-time PCR(qRT-PCR),fluorescence in situ hybridization(FISH),and miRNA sequencing.
