复方MZ-22对银屑病模型小鼠的疗效及机制初探

摘　　要：目的研究中药复方MZ-22对银屑病的改善作用及初步分子机制。方法通过银屑病皮损面积及严重程度指数(PASI)、苏木精-伊红染色、免疫组织化学染色、实时荧光定量PCR(qRT-PCR)、酶联免疫吸附试验(ELISA)、蛋白质免疫印迹分别研究MZ-22对银屑病模型小鼠银屑病样皮损、组织病理学变化、白介素-6(IL-6)和IL-17的表达、信号转导和转录激活因子3(STAT3)和磷酸化STAT3蛋白表达的影响。结果MZ-22对银屑病样皮损和组织病理学变化具有明显改善作用,可显著降低PASI评分、表皮厚度和脾脏指数;显著降低小鼠皮肤和外周血中IL-6和IL-17的表达,减轻小鼠的炎症状态;显著抑制皮肤组织中STAT3的表达和激活。结论MZ-22可能通过抑制皮肤和外周血中相关炎症因子IL-6、IL-17的表达,并抑制STAT3的表达和激活,从而缓解小鼠的银屑病症状。OBJECTIVE To explore the therapeutic effects of the traditional Chinese medicine compound formulation MZ-22 on psoriasis and its preliminary molecular mechanisms.METHODS The effects of MZ-22 on psoriasis-like skin lesions,histopathological changes,the expression of interleukin-6(IL-6)and interleukin-17(IL-17),and the protein expression of the signal transduction and activator of transcription 3(STAT3)and phosphorylated STAT3 were examined using the psoriasis area and severity index(PASI),hematoxylin-eosin staining,immunohistochemical staining,real-time fluorescent quantitative PCR,enzyme-linked immunosorbent assay,and Western blot analysis,respectively.RESULTS MZ-22 demonstrated significant ameliorative improvement effects on psoriasis-like skin lesions and histopathological changes,significantly reducing PASI scores,epidermal thickness and spleen index.It significantly reduced the expression of IL-6 and IL-17 in the skin and peripheral blood of mice,alleviating the inflammatory state of the mice.MZ-22 also significantly inhibited the expression and activation of STAT3 in skin tissue.CONCLUSION MZ-22 may alleviate the symptoms of psoriasis by inhibiting the expression of related inflammatory factors such as IL-6 and IL-17 in the skin and peripheral blood,and by inhibiting the expression and activation of STAT3.
