摘 要:目的 考察黄甲软肝颗粒对石胆酸致黄疸型肝炎大鼠模型的肝脏保护作用。方法 将60只SD大鼠随机分为正常组、模型组、阳性(茵栀黄颗粒)组及低、中、高剂量黄甲软肝颗粒组,除正常组与模型组外,其余各组均ip相应剂量的药物,连续18 d。第15日,除正常组外,其余各组连续4 d ig石胆酸玉米油溶液造模。造模24 h后,检测血清肝功能指标谷氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、总胆红素(TBIL)、胆汁酸(TBA)以及血脂指标总胆固醇(CHO)、高密度脂蛋白(HDLc)、低密度脂蛋白(LDLc)、甘油三酯(TG);观察肝脏组织病理学。结果 与模型组比较,各给药组大鼠血清中肝功能相关指标及TG的含量均有所下降,其中,黄甲软肝颗粒中、高剂量组及阳性组的ALT含量显著下降,黄甲软肝颗粒高剂量组及阳性组血清中AST、ALP、TBIL、TBA的含量显著下降,黄甲软肝颗粒中、高剂量组的TG含量明显下降;各给药组血清中CHO、HDLc、LDLc的含量无统计学差异;各给药组大鼠肝脏组织的病理损伤评分均有所下降,但无统计学差异。结论 黄甲软肝颗粒可通过降低大鼠体内ALT、AST、ALP、TBIL、TBA、TG的含量来改善石胆酸导致的黄疸型肝炎。OBJECTIVE To investigate the protective effect of Huangjia Ruangan granules(HRG) on liver in icteric hepatitis rat model induced by lithocholic acid.METHODS 60 SD rats were randomly divided into normal group, model group, positive group(Yinzhihuang granules) and low, medium and high dose of HRG groups.Except for the normal group and the model group, the other groups were given the appropriate dose of drugs for 18 d.On the 15th day, except for the normal group, all other groups were intragastrically administered with lithocholic acid corn oil solution for 4 consecutive days.After 24 h of modeling, serum liver function related indicators(ALT,AST,ALP,TBIL,TBA) and four blood lipids(CHO,HDLc, LDLc, TG) were detected.Liver histopathological analysis was conducted.RESULTS The serum levels of ALT,AST,ALP,TBIL,TBA and TG of each drug intervention group showed a decreasing trend compared to the model group.Among them, the levels of ALT in the serum of the medium and the high dose of HRG groups and the positive group decreased significantly.The levels of AST,ALP,TBIL and TBA in the serum of the high dose of HRG group and positive drug decreased significantly.The levels of TG in serum of medium and high dose of HRG group were significantly decreased.There was no significant difference in the levels of CHO,HDLc and LDLc in the serum of each group.The pathological injury score of liver tissue in each group showed a decreasing trend, but there was no significant difference.CONCLUSION HRG can improve icteric hepatitis caused by lithocholic acid by downregulating the levels of ALT,AST,ALP,TBIL,TBA and TG in rats.
