摘  要:目的研究重组贻贝粘蛋白凝胶(RMMG)的止痒效果及机制。方法研究长期使用RMMG对4-氨基吡啶致小鼠特应性皮炎的止痒作用,以苏木精-伊红和免疫组化染色法观察小鼠皮损组织的炎症变化,酶联免疫吸附测定法检测小鼠体内组织的炎症变化;以Western blotting和实时荧光定量PCR实验检测RMMG的作用机制。结果与模型组比较,RMMG组小鼠的搔抓次数明显减少,小鼠皮肤炎性浸润明显改善,皮肤组织中5-羟色胺受体2A的含量显著减少,血清中白介素-6(IL-6)、肿瘤坏死因子α(TNF-α)和组胺的水平明显降低。RMMG可能通过抑制核因子κB(NF-κB)通路发挥抗瘙痒的作用。结论RMMG可通过组胺/IL-6/TNF-α和NF-κB通路来抑制4-氨基吡啶致小鼠皮肤急性瘙痒。OBJECTIVE To study the antipruritic effect and mechanism of recombinant mussel mucin gel(RMMG).METHODS The antipruritic effect of RMMG on mice with atopic dermatitis induced by 4-aminopyridine was studied.The changes of skin inflammation in mice were observed using Hematoxylin-eosin and 5-hydroxytryptamine receptor 2A immunohistochemical staining.Tissue inflammation changes in mice were detected by enzyme-linked immunosorbent assay.Western blotting and Real-time quantitative polymerase chain reaction were used to determine the mechanism.RESULTS Compared with the model group,the scratching frequency of mice in RMMG groups was significantly reduced,the skin inflammatory infiltration showed a significant improvement,and the serum levels of IL-6,TNF-αand histamine were significantly reduced.RMMG inhibited pruritus in mice by inhibiting NF-κB pathway.CONCLUSION RMMG can inhibit acute pruritus induced by 4-aminopyridine through histamine/IL-6/TNF-αand NF-κB pathways.
