丁香对参黄散经神阙穴敷贴给药后局部药动学的影响

摘  要:目的研究参黄散配伍丁香前后各有效成分的局部药动学。方法采用UPLC-MS/MS法,将线性微透析探针植入术后肠动力障碍大鼠模型的皮下组织进行采样,采用增量法和减量法测定大黄酸、厚朴酚、吴茱萸碱、柚皮苷在皮肤中的相对损失率;动态监测4种成分在大鼠皮下组织的浓度;采用经相对损失率校正后皮肤微透析样品的浓度计算药动学参数,评价丁香对参黄散中不同成分的促渗规律。结果同等剂量下,大黄酸、厚朴酚、吴茱萸碱配伍丁香后的C_(max)分别是去丁香的1.81、2.25、1.87倍,AUC_(0-12)分别是去丁香的1.29、5.06、2.33倍,丁香对柚皮苷无促渗效果。结论微透析法可较好地采集穴位敷贴给药后的皮肤局部药动学样本,是局部药动学良好的研究手段;参黄散配伍丁香后对厚朴酚的促渗效果最强,大黄酸、吴茱萸碱次之,对柚皮苷无促渗效果。OBJECTIVE To study the local pharmacokinetics of various active ingredients of Shenhuang powder in combinations of Syzygium aromaticum.METHODS A UPLC-MS/MS method was adopted.Linear microdialysis probes were implanted into the subcutaneous tissue of a rat model of postoperative ileus.Using incremental and subtractive methods to determine the relative skin loss rates of rhein, magnolol, evodiamine, and naringin.The concentrations of four components in the subcutaneous tissue of rats were monitored dynamically.The pharmacokinetic parameters of the skin microdialysis sample concentration after relative loss rate correction were calculated to evaluate the promoting effects of S.aromaticum on the permeation of different components in Shenhuang powder.RESULTS At the same dose, the C_(max) of rhein, magnolol, evodiamine, and naringin combined with S.aromaticum was 1.81,2.25,and 1.87 times higher than that of S.aromaticum free, and AUC_(0-12) was 1.29,5.06 and 2.33 times higher than that of S.aromaticum free, respectively.S.aromaticum had no promoting effect on naringin permeability.CONCLUSION The microdialysis method proved an effective method of collecting the local pharmacokinetic samples of acupoint applications, thus representing a valuable research method for local pharmacokinetics.The combination of Shenhuang powder and S.aromaticum demonstrated the most pronounced effect on promoting the permeation of magnolol, followed by rhein and evodiamine, while exhibited no promoting effect on naringin.
