摘  要:目的研究当归的不同炮制提取物与当归补血汤抗氧化活性的谱效关系,筛选出汤剂中当归的最佳炮制方法。方法通过HPLC法建立当归补血汤不同炮制提取物的指纹图谱,测定不同炮制提取物的体外抗氧化活性;使用灰度关联法对其化学成分和体外抗氧化活性进行谱效分析。将体外抗氧化结果和不同炮制提取物中总多糖、总黄酮、总皂苷和总多酚的含量进行基于熵权法的优劣解距离法(TOPSIS)综合评价。结果美迪紫檀素、欧当归内酯A、绿原酸、洋川芎内酯H、芒柄花苷、毛蕊异黄酮、阿魏酸、洋川芎内酯I和毛蕊异黄酮葡萄糖苷对当归补血汤抗氧化作用的贡献较大;当归经过酒炙后的效果最佳。结论当归经过酒炙后,与黄芪配伍的抗氧化活性最好、有效活性成分含量最高;当归补血汤不同炮制提取物的抗氧化活性为多成分协同作用的结果。OBJECTIVE To study the spectral relationship between different processed extracts of Angelica sinensis and the antioxidant activity of Danggui Buxue decoction(DBD),and to optimize the processing method of A.sinensis in DBD.METHODS The fingerprints of the different processed extracts of DBD were established by HPLC,the in vitro antioxidant activities of the different processed extracts were determined,and the spectral analysis of their chemical composition and in vitro antioxidant activities was conducted using the gray scale correlation method.The in vitro antioxidant results and the contents of total polysaccharides,total flavonoids,total saponins and total polyphenols in the different processed extracts were subjected to entropy-weighted TOPSIS.RESULTS The results of the spectral analysis showed that medicarpin,eugenolide A,chlorogenic acid,chuanxionglactone H,mangostin,trichostatin,ferulic acid,chuanxionglactone I,and trichostatin glucoside contributed more to the antioxidant effects of the DBD.The comprehensive evaluation results indicated that A.sinensis in DBD had the best effect after wine-roasting.CONCLUSION The highest antioxidant activity and the greatest content of active ingredients were observed when A.sinensis was roasted in wine and combined with Astragalus membranaceus.The antioxidant activity of different concocted extracts of DBD was determined by the synergistic effect of multiple components.
