摘 要:为探究螺旋藻多糖(PSP)对伪狂犬病病毒(PRV)感染小鼠肝脏和肺脏氧化应激的调节作用,将120只Balb/c小鼠随机分为5组,分别为螺旋藻多糖高剂量组、中剂量组、低剂量组、PRV组和空白对照组。测定小鼠肝脏和肺脏SOD、iNOS酶活性和MDA、NO含量;HE染色观察小鼠肝脏和肺脏病理变化;免疫组化检测小鼠肝脏和肺脏中iNOS表达情况。结果显示,与PRV组相比,高剂量和中剂量能显著降低小鼠肝脏和肺脏组织iNOS酶活性及MDA、NO水平(P<0.05),显著升高SOD酶活性(P<0.05);HE染色结果显示,高、中、低剂量小鼠肝脏和肺脏病理变化有不同程度减轻;免疫组化结果显示,高、中、低剂量处理PRV感染小鼠后,肝脏和肺脏中iNOS表达有不同程度降低,高剂量组效果最佳。表明螺旋藻多糖能够减轻PRV感染小鼠肝脏和肺脏的损伤,缓解PRV感染小鼠引起的氧化应激。To investigate the regulatory effects of polysaccharide from Spirulina platensis(PSP)on oxidative stress in liver and lungs of mice infected with Pseudorabies virus(PRV),a total of 120 Balb/c mice were randomly divided into 5 groups,including high-dose group,medium-dose group,low-dose group,PRV group and blank control group.The enzyme activities of SOD and iNOS and the contents of MDA and NO in liver and lungs of mice were measured.HE staining was used to observe the histopathological changes of liver and lungs of the mice.The expression of iNOS in liver and lungs was detected by immunohistochemistry.The findings indicated that compared with PRV group,the contents of iNOS,MDA and NO in liver and lungs of the mice treated with high-dose and medium-dose PSP were significantly decreased(P<0.05),and the activity of SOD was significantly increased(P<0.05);HE staining examination revealeded that the histopathological changes of liver and lungs were alleviated to different degrees after treatment with high,medium and low-doses of PSP;The results of immunohistochemistry showed that the expression of iNOS in liver and lungs decreased to different degrees after treatment with high,medium and low-doses of PSP.High-dose PSP treatment is the most effective.The results showed that PSP can alleviate liver and lung damages in PRV-infected mice and alleviate oxidative stress caused by PRV.
