摘 要:为了解林蛙重盘吸虫的遗传多样性,对采自黑龙江省鹤岗、伊春、佳木斯、齐齐哈尔、鸡西五个地市林蛙肠道寄生的日本重盘吸虫和短肠重盘吸虫线粒体cox 1、nad 1以及核糖体ITS基因进行PCR测序和序列分析。结果表明,日本重盘吸虫3个基因部分序列的有效长度分别421 bp、905 bp和831 bp,短肠重盘吸虫3个基因部分序列有效长度分别为420 bp、739 bp和797 bp;3基因联合序列(cox1、nad1和ITS)分析不同地区50个日本重盘吸虫分离株中共发现34种单倍型,50个短肠重盘分离株中共发现46种单倍型,2种重盘吸虫各分离株均呈高单倍型多样性和低核苷酸多样性(H>0.5,π<0.005),表现出高度遗传多样性;系统发育分析显示2种重盘吸虫没有形成明显地理遗传结构;中性检验、错配分布分析结果显示,黑龙江省日本重盘吸虫可能经历过种群扩张而短肠重盘吸虫种群相对稳定。研究结果表明,黑龙江省2种重盘吸虫的遗传多样性较高,各种群间基因交流较为频繁,未形成显著的地理遗传结构。To understand the relationships among the geographic distribution of Diplodiscus japonicum and Diplodiscus mehrai in wood frogs,we amplified and analyzed the partial mitochondrial cox 1,nad 1,and ribosomal ITS genes of these two flukes isolated from intestines of wood frogs from five cities in Heilongjiang province,including Hegang,Yichun,Jiamusi,Qiqihar,and Jixi in present study.The results showed that the lengths of the three gene segments of the D.japonicum were 421bp,905bp,and 831bp,while the lengths of the three gene sequences of the D.mehrai were 420bp,739bp,and 797bp,respectively.34 haplotypes were found in 50 D.japonicum isolates and 46 haplotypes were found in 50 D.mehrai from different regions analyzed by the combined three-gene sequences(cox 1,nad 1 and ITS).Both two parasite isolates appeared to the high haplotype diversity and low nucleotide diversity(H>0.5,π<0.005)and showed high genetic diversity.A high level of genetic diversity was demonstrated among the different isolates of the two parasites from different regions.Phylogenetic analyses indicated that D.japonicum and D.mehrai did not form obvious distinct geographic genetic structures.The results of the neutrality tests and mismatch distribution analysis revealed that D.japonicum from Heilongjiang province may have experienced population expansion,while D.mehrai population remained relatively stable.The results of present study showed that the genetic diversity of D.japonicum and D.mehrai in Heilongjiang province was high,with frequent gene exchanges,but no significant geographic genetic structure was formed.
