摘 要:通过小鼠急性毒性试验和大鼠亚慢性毒性试验,对柴茵护肝合剂进行安全性评价。在急性毒性试验中,对小鼠进行一次性灌胃给药,测定柴茵护肝合剂的半数致死量和最大耐受量;在亚慢性毒性试验中,将80只大鼠随机分为高、中、低剂量组(20、10、5 g/kg)和空白对照组,连续给药28 d及停药后7 d,每组随机取10只大鼠,进行血液生理、生化指标的测定和病理学组织学检查。结果显示,小鼠急性毒性试验无法测得LD 50,最大给药量为20 g/kg体重。大鼠亚慢性毒性试验结果显示,高、中、低剂量的柴茵护肝合剂对大鼠临床体征、生长性能和血液生理生化无不良影响,剖检和组织病理学检查未见脏器有明显病变。表明柴茵护肝合剂安全性好,无毒副作用。This study was carried out to evaluate the safety of Chaiyinhugan mixture through acute toxicity test in mice and subchronic toxicity test in rats.In the acute toxicity test,a single oral dose was administered to mice to determine the median lethal dose(LD 50)and maximum tolerance dose of Chaiyinhugan mixture.In the subchronic toxicity test,80 rats were randomly divided into high,medium,low-dose groups(20 g/kg,10 g/kg,5 g/kg)and a blank control group,20 rats each group.The animals were continuously administered with the mixture for 28 days.After the treatment,10 rats from each group were randomly selected for blood physiological and biochemical index measurements,as well as histopathological examination.Clinical observation was conducted for an additional 7 days after drug withdrawal.Blood physiological,biochemical and organ index measurements,as well as histopathological examinations,were performed at the end of the observation period.The results showed that the LD 50 of Chaiyinhugan mixture could not be obtained in the acute toxicity test,and the maximum administration dose was 20 g/kg·bw.In the subchronic toxicity test,the mixture showed no adverse effects on clinical signs,growth performance,blood physiological or biochemical parameters in the different dose groups of rats.Additionally,no significant organ lesions were observed in the dissection and histopathological examination.The result showed that Chaiyinhugan mixture has a high safety and is non-toxic.
