摘  要:程序性死亡受体-1(programmed death-1,PD-1)是一种免疫抑制性受体,通过与表现于特定淋巴细胞表面的PD-L1配体结合,触发淋巴细胞死亡,进而抑制免疫反应。在某些慢性病毒感染及肿瘤等疾病的进程中,PD-1及其配体的表达量及转录水平显著升高,这导致免疫系统中T细胞的激活受阻,免疫功能随之受损。探索干预PD-1通路的目的是恢复T细胞的活性。论文梳理了近年来关于PD-1/PD-L1通路的研究进展,特别是单克隆抗体类拮抗剂、小分子肽类拮抗剂及小分子非肽类抑制剂等领域的最新发现,为免疫联合治疗提供新的策略和方法,旨在逆转动物和人体内的免疫抑制状态,开拓联合免疫治疗的新前景。PD-1 is an immunosuppressive receptor that induces lymphocyte death by binding with its ligand PD-L1,displayed on certain lymphocytes'surfaces,thereby suppressing the immune response.Extensive research in recent years has revealed that in conditions such as chronic viral infections and cancers,there is a significant increase in the expression and transcription levels of PD-1 and its ligand.This results in the inhibition of T cell activation and subsequent impairment of immune function.Given these findings,researchers have begun to explore strategies to intervene in the PD-1 pathway with the aim of restoring T cell activity and enhancing immune responses.This review compiles recent advances in research on the PD-1/PD-L1 pathway,with a particular focus on monoclonal antibodies as antagonists,small-molecule peptide antagonists,and small-molecule non-peptide inhibitors.These discoveries offer new strategies and methods for combination immune therapy aimed at reversing immune suppression in both animals and humans,paving new avenues for immune therapeutic treatments.
