摘 要:为比较DHAV-3结构蛋白免疫保护性差异,利用原核表达系统表达重组蛋白,制备鸭抗DHAV-3-VP0、-VP3、-VP1多克隆抗体,并测定其中和效价;以制备的r-DHAV-3结构蛋白为免疫原免疫SPF鸭后攻毒,通过保护率、组织病理学分析及病毒载量,评价DHAV-3的3种结构蛋白免疫原性及免疫保护性差异。结果表明:鸭抗DHAV-3-VP0、-VP3、-VP1多克隆抗体ELISA效价分别为1:102 400、1:51 200、1:204 800;中和效价以鸭抗DHAV-3-VP1多克隆抗体最高为1:178,VP3次之为1:149,VP0中和效价最低为1:53;rDHAV-3-optiVP1免疫组保护率为60%;r-DHAV-3-VP0、r-DHAV-3-VP3免疫组保护率为40%。所有免疫组肝脏组织病理损伤程度及肝脏、脾脏病毒载量均显著低于对照组。DHAV-3的3种结构蛋白免疫保护性以VP1最强,VP3、VP0次之。To compare the differences in the immunoprotective properties of DHAV-3 structural proteins,recombinant proteins were expressed using a prokaryotic expression system to prepare duck anti DHAV-3-VP0/-VP3/-VP1 polyclonal antibodies,and their titers were determined.Using the prepared r-DHAV-3 structural protein as the immunogen to immunize SPF ducks,the immunogenicity and immunoprotective differences of the three structural proteins of DHAV-3 were evaluated through protection rate,histopathological analysis,and viral load.The results showed that the ELISA titers of duck anti DHAV-3-VP0/-VP3/-VP1 polyclonal antibodies were 1102400,151200 and 1204800,respectively.The neutralizing potency of duck anti DHAV-3-VP1 polyclonal antibody was the highest at 1178,followed by VP3 at 1149,and VP0 had the lowest neutralizing potency at 153.The protection rate of the r-DHAV-3-opiVP1 immune group was 60%.The protection rate of the r-DHAV-3-VP0 and r-DHAV-3-VP3 immune groups was 40%.The degree of pathological damage to liver tissue and the viral load of liver and spleen in all the immune groups were significantly lower than those in the control group.It indicated that VP1 had the strongest immunoprotective activity among the three structural proteins of DHAV-3,followed by VP3 and VP0.
