摘 要:基质金属蛋白酶23B(MMP23B)是基质金属蛋白酶家族成员,在胚胎发育、生殖和组织重塑、细胞增殖与分化、血管生成等过程中发挥作用。为研究MMP23B是否参与奶牛乳腺中乳脂合成调节,研究构建牛MMP23B表达和敲降载体,采用基因过表达和干扰方法改变奶牛乳腺上皮细胞MMP23B表达水平,检测MMP23B表达变化对乳脂合成相关基因及细胞中甘油三酯含量的影响。荧光定量PCR结果表明:MMP23B过表达或敲降显著提高或降低奶牛乳腺上皮细胞MMP23B、FASN、ACC、SCD1 mRNA表达。Western blot结果表明,MMP23B过表达显著提高奶牛乳腺上皮细胞MMP23B和FASN蛋白表达;MMP23B敲降后MMP23B和FASN蛋白表达降低。同时MMP23B过表达显著提高奶牛乳腺上皮细胞甘油三酯质量分数;当MMP23B敲降后甘油三酯质量分数降低。综上,MMP23B可正向调节乳腺上皮细胞甘油三酯合成。Extracellular matrix 23B(MMP23B)is a member of the matrix metalloproteinase family and plays a role in embryonic development,reproduction and tissue remodeling,cell proliferation and differentiation,and angiogenesis.In order to investigate whether MMP23B was involved in the regulation of milk fat synthesis in the mammary glands of dairy cows,the bovine MMP23B expression vector and knock-down vector were first constructed,and the expression levels of MMP23B in mammary epithelial cells of dairy cows were the effects of MMP23B expression on the genes related to milk fat synthesis and the content of triglyceride in cells were examined when MMP23B was overexpressed and knocked down.The results of qPCR showed that overexpression or knockdown of MMP23B significantly increased or decreased the mRNA expression of MMP23B,FASN,ACC and SCD1 in mammary epithelial cells of dairy cows.The results of Western blot showed that overexpression of MMP23B significantly increased the expression of MMP23B and FASN proteins in mammary epithelial cells of dairy cows,and decreased the expression of MMP23B and FASN proteins after knockdown of MMP23B.Overexpression of MMP23B significantly increased the content of triglyceride and decreased the content of triglyceride when MMP23B was knocked down.These results suggested that MMP23B could positively regulate triglyceride synthesis in mammary epithelial cells of dairy cows.
