摘 要:目的:探讨三七总皂苷乳膏对皮肤缺损模型小鼠的治疗作用。方法:60只SPF级健康雌性小鼠,随机分为模型组、阳性对照组(重组人表皮生长因子凝胶)、空白乳膏基质组、三七乳膏组,每组15只。在小鼠背部中线处剪去直径为1 cm的圆形皮肤直达筋膜层,建立全层皮肤缺损模型。造模2 h后给药,每日更换1次药物,连续干预21 d。观察小鼠创面愈合情况,造模后3 d、7 d、14 d计算创面愈合率;HE及Masson染色观察创面病理情况;ELISA检测血清中TNF-α、IL-6以及VEGFA、STAT3、HSP90AA1的表达水平。结果:随着用药时间的推移,三七乳膏组和阳性对照组的创面变化更为显著,愈合速率明显加快(P<0.05);HE染色、Masson染色结果显示,三七乳膏组和阳性对照组的炎性细胞增殖相较于其他两组显著降低。小鼠创面新生毛细血管、成纤维细胞等数量在各组中均呈现不同程度的增长,尤其在三七乳膏组和阳性对照组增幅明显;三七乳膏能够减少血清中炎症因子IL-6、TNF-α的含量(P<0.05,P<0.01),同时,还能调节小鼠皮肤组织中STAT3蛋白的表达,上调VEGFA和HSP90AA1的表达水平(P<0.05,P<0.01)。结论:三七总皂苷对全层皮肤缺损模型小鼠的皮肤创面愈合有促进作用。Objective:To investigate the therapeutic effects of Panax Notoginseng total saponins ointment on skin defect models in mice.Methods:Sixty SPF-grade healthy female mice were randomly divided into the model group,positive control group(recombinant human epidermal growth factor gel),blank ointment matrix group,and Panax Notoginseng ointment group,with 15 mice in each group.A full-thickness skin defect model was established by excising a 1 cm circular piece of skin down to the fascia along the midline of the mices backs.After 2 hours of modeling,treatments were applied,with medication replaced once daily for 21 consecutive days.Wound healing was observed,and the healing rates were calculated on days 3,7,and 14 after modeling.Hematoxylin-eosin(HE)and Masson staining were performed to observe the pathological changes of the wound.Enzyme-linked immunosorbent assay(ELISA)was used to detect serum levels of TNF-α,IL-6,VEGFA,STAT3,and HSP90AA1.Results:Over time,significant changes in the wound were observed in both the Panax Notoginseng ointment group and the positive control group,with a notably faster healing rate(P<0.05).HE and Masson staining results showed that the proliferation of inflammatory cells was significantly reduced in the Panax Notoginseng ointment group and the positive control group compared to the other two groups(P<0.05).The number of new capillaries and fibroblasts in the wound tissue increased to varying extents in all groups,especially in the Panax Notoginseng ointment group and the positive control group,with a significant increase(P<0.05).Panax Notoginseng ointment reduced the levels of inflammatory factors IL-6 and TNF-αin the serum(P<0.05)and modulated the expression of STAT3 protein in the skin tissue,upregulating the expression levels of VEGFA and HSP90AA1(P<0.05).Conclusion:Panax Notoginseng total saponins promote skin wound healing in a full-thickness skin defect model in mice.
