摘 要:为建立施氏鲟(Acipenser schrencki)单性育种技术,本研究采用组织病理学、生化指标及高通量测序技术探究不同浓度的外源性17ɑ-甲基睾酮(MT)对施氏鲟性腺组织形态、类固醇激素、转录组表达模式及性别决定基因表达水平的影响。选取初始体质量为(5.12±0.12)g的3月龄施氏鲟,使用MT对其进行性逆转诱导。所有试验鱼随机分为四组:对照组(M0),10 mg/mL MT处理组(M1),30 mg/mL MT处理组(M2),50 mg/mL MT处理组(M3)。每组120尾个体,试验周期为3个月。结果表明,MT投喂60 d时,卵原细胞逐渐空泡化,各处理组出现精卵并存现象,投喂90 d时,观察到性腺结构中有精原细胞存在。此外,不同剂量MT投喂后雌二醇含量显著低于对照组,而睾酮含量则无明显差异。转录组结果显示,养分库活性、激素活性、雄激素生物合成和皮质醇生成相关的GO条目被显著富集。基于KEGG富集分析发现,皮质醇合成与分泌、卵巢类固醇生成和神经活性配体-受体相互作用等通路显著富集。实时荧光定量PCR结果显示,FOXL2与cyp19ala表达量显著下调,Dmrt1表达量显著上调。研究结果表明,不同浓度MT处理下施氏鲟性腺组织结构、类固醇激素、基因表达及性别决定相关基因的表达水平均受到显著影响。The present study was carried out to investigate the effects of different concentrations of exogenous 17ɑ-methyltestosterone(MT)on the morphology of gonadal tissues,steroid hormones,expression patterns of the transcriptome,and the expression levels of sex-determining genes of Acipenser schrencki using histopathological and biochemical indexes as well as high-throughput sequencing techniques to establish a unisexual breeding technique for A.schrencki.A cohort of three-month-old Acipenser schrencki with an initial body weight of 5.12±0.12 g was selected for sex reversal induction using MT.The test fish were randomly divided into four groups as control group(M0),10 mg/mL MT-treated group(M1),30 mg/mL MT-treated group(M2)and 50 mg/mL MT-treated group(M3).Each group consisted of 120 individuals,and the experimental period was three months.The results demonstrated that at 60 days of MT administration,the oogonia exhibited gradual vacuolization,and the coexistence of spermatozoa was observed in all treatment groups.At 90 days of MT administration,spermatogonia in the gonadal structure was evident.Furthermore,the oestradiol content was significantly lower than that of the control group after administration of different MT doses.In contrast,the testosterone content was not significantly different.The transcriptome results demonstrated that GO entries associated with nutrient pool activity,hormone activity,androgen biosynthesis,and cortisol production exhibited a notable enrichment.The KEGG-based enrichment analysis revealed significant enrichment of pathways for cortisol synthesis and secretion,ovarian steroidogenesis,and neuroactive ligand-receptor interactions.The real-time fluorescence quantitative PCR results demonstrated a significant down-regulation in the expression of FOXL2 and cyp19ala,while the expression of Dmrt1 was found to be significantly up-regulated.The results indicated that different concentrations of MT treatment in A.schrencki significantly affected the gonadal tissue structure,steroid hormones,gene
