摘 要:类风湿关节炎(RA)是一种病因未明的慢性全身性自身免疫性疾病,以关节滑膜炎症、软骨破坏及全身并发症为特征,显著增加致残率和死亡率。近年来,多组学技术的整合分析为解析RA的复杂发病机制提供了全新视角。多组学技术通过整合遗传变异、表观调控、蛋白质动态及代谢通路,系统性阐述了RA的分子特征,为发现潜在的生物标志物和开发靶向治疗策略提供了理论依据。本文系统综述了基因组学、表观遗传学、蛋白质组学及代谢组学在RA研究中的进展。未来需进一步融合多组学数据,建立基于分子特征的RA分型体系,并推动转化医学研究,以解决临床实践中的挑战。Rheumatoid arthritis(RA)is a chronic systemic autoimmune disease of unknown etiology,characterized by synovial inflammation,cartilage destruction,and systemic complications,which significantly increase disability and mortality rates.In recent years,the integration of multi-omics technologies has provided novel perspectives for elucidating the complex pathogenesis of RA.By consolidating genetic variations,epigenetic regulation,protein dynamics,and metabolic pathways,multi-omics technologies have systematically elucidated the molecular characteristics of RA,providing a theoretical foundation for identifying potential biomarkers and developing targeted therapeutic strategies.This article systematically reviews the progress in genomics,epigenetics,proteomics,and metabolomics in RA research.Future efforts should focus on further integrating multi-omics data to establish a classification system for RA based on molecular characteristics and to promote translational medical research,thereby addressing challenges in clinical practice.
